K-ras基因突变在肺腺癌发生及诊治中的新认识

摘要:
K-ras基因突变常见于肺腺癌。研究认为吸烟是促发K-ras基因突变的重要因素,K-ras基因突变对肺腺癌亚型的演变及肺腺癌的预后产生影响。近年来K-ras基因的检测手段不断创新,大大提高了对肿瘤成分较少的病理组织的突变基因的检测率。目前认为K-ras基因突变会对抗表皮生长因子受体(EGFR)药物产生抗药性,并对化疗疗效产生一定的影响。因此,关于K-ras基因突变的诊断与靶向治疗的关系已成为国内外研究的热点。本文通过对近些年来有关K-ras基因突变与肺腺癌文献进行回顾,对伴有K-ras基因突变的肺腺癌的发生、诊断和治疗做一综述。

Abstract:
K-ras gene mutation is commonly seen in lung adenocarcinoma. Cigarette smoking is thought to be an important reason for K-ras gene mutation. Studies have reported that K-ras mutations can influence the subtyping and prognosis of lung adenocarcinoma. Recently, the techniques to detect K-ras mutation have been continuously improved, greatly promoting the detection sensitivity of K-ras mutations, even in tissues with small amount of tumor component. Emerging data showed that K-ras mutation can induce primary resistance of lung adenocarcinoma to tyrosine kinase inhibitors (TKIs) and decrease the efficacy of chemotherapy. Therefore, K-ras mutation has attracted considerable attention as a target for diagnosis and anticancer therapy. In this review, we summarize recent studies on K-ras mutations in lung adenocarcinoma, and discuss the development, diagnosis, and treatment of lung adenocarcinoma.
