    目的:探讨钾通道激动剂在哮喘防治中的作用。方法:测定哮喘动物模型气道阻力、肺泡灌洗液细胞计数及病理检查,观察钾通道激动剂BRL 55834对气道阻力及气道炎性细胞浸润的影响,并与对照剂二甲亚砜(DMSO)、氨茶碱、BRL 38227及维拉帕米比较。结果:预先静注BRL 55834可抑制卵蛋白诱发致敏豚鼠气道痉挛,减轻气道炎性细胞浸润,对血压无明显影响;BRL 38227及氨茶碱均能抑制炎性细胞浸润,氨茶碱还能抑制卵蛋白诱发的气道痉挛,但BRL 38227可降低豚鼠血压;维拉帕米对卵蛋白诱发的气道痉挛及炎性细胞浸润均无抑制作用。结论:钾通道激动剂BRL 55834具有气道相对选择性,抑制抗原所致气道阻力升高及气道炎性细胞浸润,且对心血管系统无明显影响,具有临床应用前景。

    Abstract:

    Objective: To study the role of potassium channel activator in treatment of asthma. Methods: Airway insufflation pressure measurement(AIP), bronchial alveolar lavage fluid(BALF) cells count and pathological analysis were employed to detect airway resistance variation and airway inflammatory cells infiltration following inhalation of ovalbumin. Results: BRL 55834(8 μg/kg) inhibited not only ovalbumin induced AIP increase but also inflammatory cells infiltration in sensitized guinea-pigs, it did not lower blood pressure. By contrast to BRL 55834, BRL 38227 (200 μg/kg) and aminophylline(25 mg/kg) inhibited ovalbumin induced inflammatory cells infiltration, but BRL 38227 lowered blood pressure markedly. Verapamil (0.5 mg/kg) had no effects on AIP increasing and inflammatory cells infiltration. Conclusions: BRL 55834, a selective potassium channel activator, not only decreases airway resistance, but also inhibits inflammatory cells infiltration, and this is beneficial for treatment of asthma.
