Abstract:Objective:To study systematically the role of skeletal muscle cells in synthesizing, processing and secreting recombinant proteins in vitro. Methods: Primary murine myoblasts from SCID mice (SCID-MB) and C2C12, a mouse myoblast cell line, were transfected with muscle specific (pdLMe4bAhⅨm) or non-specific (LⅨSN) vectors encoding human factor Ⅸ (FⅨ). The capacities of the transgenic cells in producing recombinant FⅨ were compared in the levels of intracellular and secreted FⅨ protein as well as FⅨ mRNA in primary muscle cells and C2C12 cells. Results: Both primary muscle cells and C2C12 cells can efficiently translate and process FⅨ protein. Myotubes derived from the SCID-MB produced-2000 ng FⅨ.(106 cells)-1.(24 h)-1 in culture with specific activity of 95%?103%. C2C12 cells can synthesize and process recombinant FⅨ as efficiently as primary muscle cells, but their secretion ability is less efficient in comparison with primary cells. Conclusion: The primary cells should be used in gene therapy related research projects and care should be taken while explaining the results derived from established cell lines.

Abstract:Objective: To obtain the homozygote of HBV transgenic mice with unique integrated site by genetic breeding. Methods: The positive transgenic mice were mated with the normal mice of the same line. And the target gene integration ratio were tested by PCR, Dot-blotting and Southern-blotting, and when the integration ratio was almost 50%, the positive mice were mated with inbreeding. Results: The 7 generations of HBV transgenic mice were obtained, and the homozygote of HBV transgenic mouse was obtained. Conclusion: The HBV DNA can be transmitted in the transgenic mice stably, and the homozygote of HBV transgenic mouse was obtained.

Abstract:Objective:To study the mechanism of neutral proteinases in degenerated cervical disc. Methods: Twenty white New Zealand rabbits were separated into 2 groups: model group and control group, each group consisted of 2 subgroup, each subgroup had 5 rabbits. The experimental model of degenerated cervical disc in the rabbit was established by resection of cervical supraspinous and interspinous ligaments and detachment of posterior paravertebral muscles from the cervical vertebrae. Rabbits in 3 month groups and in 8 month groups were killed 3 months and 8 months after operation respectively. The cervical disc tissues in all 4 groups were assayed to detect collagenase and serine proteinase activities by biochemical methods. Results: Collagenase and serine proteinase activities in discs from model subgroups were obviously higher than those of control subgroups. Collagenase and serine proteinase activities in discs from model subgroup in 3 month were higher than those of model subgroup in 8 month. Conclusion: Collagenase and serine proteinase activities in degenerated cervical discs are increased obviously, and their increase extents are not related to the extent of cervical disc degeneration. The results suggest that the collagenase and serine proteinase play an important role in early stage of the cervical disc degeneration.

Abstract:Objective: To investigate TTV infection among blood donors with normal ALT, the transmission of TTV, and the association between TTV and hepatitis. Methods: Plasma samples from donors with normal ALT were detected for TTV DNA using PCR method, the PCR products (TTVD026) were sequenced directly. Sequence homology analysis was conducted by Internet BLAST. Results: TTV DNA was detected in 20 of 120 donors with normal ALT, the positive rate was 16.7%. Compared with TA278, GH1, TTVCHN1 and TTVCHN2, the nucleotide identity of TTVD026 were 99%, 99%, 99%and 89%, respectively. Conclusion: TTV infection exists in blood donors with normal ALT. TTVD026 might belong to the same genotype as TA278, GH1, TTVCHN1 and TTVCHN2, which indicate that blood transfusion may play an important role in the transmission of TTV.

Abstract:Objective::To increase tumor immunogenicity by enhancing gene expression of TNF,IFN,MHC Ⅰ molecule , adenoviral vectors containing HuTNFα and IFNβ cDNA driven by HLA-B7 inducible promoter and CMV promoter were constructed, respectively. Methods and Results:The 2 fragments HLA-B7 promoter-TNF and IRES-IFN were isolated from and pGL3B7TNF and pIRIF and insert to pBluescript Ⅱ Sk (+) to generate pBLB7TNIRIF.TNFα-IRES-IFNβ was isolated and ligated to AdCMVLink1 to generate AdCMVTNIRIF(+), in which gene transcription was driven by CMV promoter.B7pro-TNFα-IRES-IFNβ was isolated and ligated to AdBglⅡto generate AdB7TNIRIF(-),in which gene expression was driven by HLA-B7 promoter. Conclusion:The constructed vectors can be a new route for enlarging tumor immunogenicity.

Abstract:Objective:To evaluate the effect of herpes simplex virus thymidine kinase gene and ganciclovir system(HSV-TK/GCV) on humoral and cellular immunological functions in malignant glioma patients.Methods:Twelve patients of malignant glioma were treated with the HSV-TK/GCV system. Peripheral blood and cerebral spinal fluid(CSF) were drawn before, during and afer treatment. Flow cytometry was used to analyze the T lymphocyte subclone of peripheral blood samples. Serum and CSF immunoglobins were quantified through turbidimetry. Results: CD4+T lymphocyte was found elevated during the treatment; NK level fell during the treatment, but rose after treatment. After treatment, cytotoxic lymphocytes(CTL) level was significantly higher than before treatment. Serum IgM and CSF IgG, IgA and IgM levels were seen higher after treatment. Conclusion: HSV-TK/GCV system can stimulate the humoral and cellular immunological functions in malignant glioma patients.

Abstract:Objective: To evaluate the antiinflammatory and antioxidant effects of the extracts of Scrophularia ningpoensis Hemsl. (SN) in vivo and in vitro. Methods: (1) Carrageenan and cobra venom induced rat paw edema; (2) Fe2+/cystine induced rat liver microsomal lipid peroxidation and (3) 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH) induced oxidative hemolysis were used to study the effects of the extracts from SN. Results: The crude extract of SN had more potent antiinflammatory effect in vivo. At a dose corresponding to the quantity contained in the crude extracts, the phenylpropanoid glycosides had potent antioxidant effects, while the iridoid glycosides of SN had lower antioxidant activities. Conclusion: The antiinflammatory activity of SN appears to have some relationship with the antioxidant effect of phenylpropanoid glycosides.

Abstract:Objective::To study the toxic and pharmocodynamic effects of sustained release preparation of norcantharidin (NCTD). Methods: Poloxamer 407 (P407) gel was used as a sustained-release vehicle for topical administration of NCTD. The toxicity of different preparations of NCTD in mice were observed. The antitumor effects of NCTD or NCTD and P407 (NCTD/P407) on SD rats implanted with W256 carcinoma were also studied. Results: (1) The sustained release preparation of norcantharidin in Poloxamer 407 (NCTD/P407) might stay in the liver at least for 4 h after injection. (2) The toxicity of the sustained release preparation of NCTD in P407 gel was lower than that of free NCTD. (3) There were significant slower tumor growth, more extensive tumor necrosis and longer survival in SD rats treated with NCTD/P407 than those treated with free NCTD. Conclusion: The NCTD in P407 gel is less toxic and have more tumoricidal effects than the equivulent dose of free NCTD, mainly because NCTD in P407 may stay in the injecting location and act with the tumor cells for a longer time.

Abstract:Objective: To investigate the regulating role of Nei Yi Fang (NYF) on cytokines in endometriosis. Methods: According to Cummings AM method, animal models with endometriosis was established. IL-6 and IL-10 levels were determined by double antibody sandwich ELISA. Results: The peritoneal macrophage amounts and the production of IL-6 by LPS-activated peritoneal macrophages were remarkably higher in untreated group than that in the control group. The weights of endometriotic tissue and peritoneal macrophage amounts were remarkably lower in treated group compared with untreated group, and the levels of IL-6 by LPS-activated peritoneal macrophages were significantly reduced. The LPS-stimulated IL-10 secretion in peritoneal macrophages from untreated group was significantly lower compared with control group and treated group. Conclusion: The lost balance of IL-6 and IL-10 levels in endometriosis may be related to the pathogenesis of the disease. The treated mechanism of NYF was mediated by restraint of IL-6 composition, and promotion of IL-10 secretion.

Abstract:Objective:To study the incidence, the antibiotics sensitivity and clinical characteristics of respiratory infection to H.influenzae in chronic obstructive pulmonary disease (COPD). Methods: Improved Columbia chocolate agar was used for the primary isolation of H.influenzae from sputums. The sensitivities of 20 antibiotics to H.influenzae were determined in vitro by Kirby-Bauer method and MICs of 12 antibiotics were determined by broth microdilution method. The clinical characteristics of respiratory infection to H.influenzae in COPD patients were analyzed. Results: Among 421 cases of COPD with acute acceleration, 229 strains of bacterium were isolated, 45(19.7%) were H.influenzae. The third generation of cephalosporins and the quinotones were most active against H.influenzae. The resistant rate against H.influenzae of Ampicillin was 22.2%.Conclusion:The infection due to H.influenzae is one of the most important agents leading to acute acceleration in COPD. The clinical characteristics of respiratory infection due to H.influenzae in COPD are no special.

Abstract:Objective: To study the protective effect of neutral mucopolysaccharide (NM) on RBC membrane during ischemia reperfusion brain damage in gerbil. Methods: The functional changes of red blood cell immune adherence (RCIA) and the content of red blood cell superoxidate dismustase (RBC-SOD), red blood cell malonialdehyde (RBC-MDA) were observed by means of cerebral ischemia reperfusion in gerbil. Results: NM recovered the function of RCIA; increased RBC-SOD; decreased RBC-MDA. Conclusion: NM regulates the function of RCIA by antioxygenic effection, and has the protective effect on RBC membrane during ischemia reperfusion brain damage.

Abstract:Objective: To study the influence of electret field on fibroblast cell cycle and to probe its mechanism. Methods: The effect of teflon (PTFE) electret on the parameter of fibroblast cell cycle with flow cytometry in 2 dimentional dot diagram was studied. Results: (1) After 24 h action of negative PTFE, the cell number in S phase increased. Negative electret could improve cell growth. (2) Negative electret not only enhanced the growth of cells, but also enhanced the apoptosis of cell. (3) After 24 h action of positive electret on fibroblast, the cell number in G1 phase increased, while the cell number decreased in S and G2 phase. Conclusion: Negative electret may improve cell growth. Positive electret could block some cell in G1 phase. So it may restrain the cell growth of fibroblast.

Abstract:Objective::To investigate the therapeutic effects of somatostatin(SS) and oxymatrine(Oxy) on experimental pancreatitis and their mechanisms. Methods: Acute experimental pancreatitis rats were induced with supramaximal dosage of caerulein injected subcutaneously. The treatment group administered with SS intravenously and Oxy i.p.,and related parameters were examined. Results: Compared with acute pancreatitis group, pancreatic mass, edema, inflammatory cell infiltration and serum amylase were significantly reduced in SS or Oxy treatment group.Besides,the inflammatory cytokines in serum or pancreatic tissue were both significantly decreased(P<0.01). Conclusion:SS and Oxy have beneficial effects on treatment of experimental acute pancreatitis. The mechanism maybe related to inhibiting the releasing and activity of inflammatory cytokines

Abstract:Objective: To observe the mechanisms of imported highly purifi ed As2O3 inhibiting human gastric carcinoma cell lines. Methods: A wide range concentration of As2O3 (0.1-2.0 μmol/L) were cultured w ith gastric carcinoma cell lines SGC7901 and MKN45 with different time, the n, the number of active cells, cell morphology, cell-DNA content distribution were evaluated. NIH3T3 cells were as controls. Results: 1.0-2 .0 μm ol/L As2O3 treated cells presented some typical features of apoptosis such a s intact cell membrane, chromatin condensation, nucleic fragmentation and apopto tic body formation, The analysis of flow cytometery showed that sub-G1 cells immerged before the G1 phase, and show ed a positive correlation with the culture period and concentration of As2O3 . Agrose electrophoresis showed a specific DNA apoptotic ladder. Conclu sion: As2O3 can inhibit human gastric carcinoma cells, the induction of apoptosis is a very important mechanism of As2O3 to treat gastric carcinoma cells.

Abstract:Objective:To assess and improve the therapeutic effect of interstitial brachytherapy. Methods: A hundred and seventy-three patients with deep brain tumors were treated with interstitial radio therapy through 3 different methods and at 3 different doses in different phases, they were followed up in different periods. Results: No operative death occured in all treated patients. Follow-up of 5-82 months revealed that 3 years survival rate of astrocyte tumors patients of Ⅰ?Ⅱ degrees was 54.5%, the survival rate of 5 years was 38.6%; one year with Ⅲ?Ⅳ degrees was 11.6%, two years was 72.0%, three years was 16.2%. The result of radiology follow-up survey indicated: fifteen cases with tumors completely disappeared (8.7%); 29 cases with tumors dimension shrinking over 80%(16.8%); 81 cases with shrinking 30%?50%(46.8%). Tumors of complete disapperance were intracranial malignant lymphoma, pineal cell tumors, multiform gliblastoma and metastatic tumors etc. Conclusion: To decrease the complications of radiation, improve the effect of treatment, different locations and properties of intracranial tumors should be treated with different radiation doses. The method of ultra-segmentation plus extra-radiation are better.

Abstract:Objective: To explore X-knife stereotactic radiosurgery of intracranial lesions. Methods: We had treated 226 patients of intracranial lesions using X-knife radiosurgery. Results: Two hundred and twenty-six patients with intracranial lesion were followed up by neurological images between 1 month and 3 years. Intracranial lesions disappeared in 38 cases (28.4%),shrinked in 45 (33.6%), and unchanged in 35 (26.1%), grew in 11 (11.9%). Conclusion: X-knife has different treatment result for intracranial lesions, brain metastasis is good using X-knife radiosurgery.

Abstract:Objective: To prepare Gaoyuan skin care cream (O/W) for plateau environment. Methods: Gaoyuan skin care cream (O/W) was specially prepared with OP,silicon,Salol,VitE,TiO2, etc. Test of antiultraviolet were performed using photoxic reaction model of chloromazine. Results: Stabilization test demonstrated that the cream had an excellent physical and chemical stability. The effect of antiultraviolet of Gaoyuan skin care cream was higher than that of control group (P<0.01). Conclusion: The effect of antiultraviolet is excellent and it is a safe and effective cream.

Abstract:Objective: To compare the pharmacokinetics and bioavailability of nicardipine hydrochloride sustained-release pellets (NIC-SP) with common market tablets and imported sustained-release capsules (Perdipine LA). Methods: Latin test was employed in the single oral administration of NIC-SP, common market tablets and Perdipine LA with 6 volunteers. The relative bioavailabilities of NIC-SP against market tablet and Perdipine LA were calculated. Results: The bioavailability of NIC-SP against Perdipine LA is 116.09%. The bioavailabilities of NIC-SP and Perdipine LA against market tablet is 86.53%and 74.54%, respectively. There were significant differences between AUC, MRT of NIC-SP and Perdipine LA (P<0.05), while tmax,cmax,ke and ka of them did not show significant difference(P>0.05). Conclusion: The sustained effect of NIC-SP is better than the Perdipine LA in vivo.

Abstract:Objective: To study the antifungal activity of the new triazole compounds against candida species. Methods: The MICs of new triazole compounds were determined in in vitro antifungal tests to evaluate the activities of title compounds. Results: All new triazole compounds had activity against candida species, especially high activity against candida albicans. Compound 12 exhibited remarkably high efficacy and was 256 times more active than fluconazole against candida albicans. Compound 17 was the most effective agent against fluconazole-resistant candida albicans and was 16 times more active than fluconazole. Conclusion: The 2 compounds are worth advanced studying as potential drugs.

Abstract:investigate the sinus and atrioventricular node functions in patients with rheumatic heart disease combined with chronic atrial fibrillation (AF).Methods:Thirty rheumatic heart disease patients with chronic AF underwent epicardial mapping during valvular operation.The sinus and atrioventricular node functions were assessed after cardioversion.Results:Twenty-eight patients had their sinus rhythm restored after undergoing electrical cardioversion.The success rate was 94%.None of the 28 patients showed sinus and atrioventricular node hypofunction.The mean SNRT was (840.7±199.1) ms;mean SNRTc was (231.8±103.1) ms,all the atrioventricular Wenckebach points were over 130 min-1 with a mean value of (327.1±37.1) ms,all the atrioventricular conduction 2∶1 block were over 150 min-1.Conclusion:After the electrical cardioversion,the sinus and atrioventricular node functions of most rheumatic heart disease patients with chronic AF stay within the normal range.The results suggested that these patients should be given the opportunity to undergo cardioversion promptly.

Abstract:Objective: To investigate collagen remodeling in infarct zone(IZ) and non-infarct zone(NIZ) in rat model of myocardial infarction. Methods:Large myocardial infarction of left ventricle was produced in rats by ligation of left coronary artery.Collagen contents were measured with the chloramine T method and collagen type Ⅰ/Ⅲ ratios were analysed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.The changes of collagen content and collagen type Ⅰ/Ⅲ ratio in IZ and NIZ were sequentially observed at 1,3, 7,14, and 42 d after myocardial infarction. Results: Collagen contents in IZ and NIZ increased gradually with time and were higher at 3, 7, 14 and 42 d than those in sham group at corresponding time (P<0.05). Compared with sham group,collagen type Ⅰ/Ⅲ ratio in IZ was lower at 3 d, and increased significantly after 7 d(P<0.01). The collagen type Ⅰ/Ⅲ ratio in NIZ was higher at 14 d (P<0.01) and 42 d (P<0.05). Conclusion: Collagen remodelling occured in both infarct zone and non-infarct zone after myocardial infarction,which may play an important role in the process of left ventricular remodelling after myocardial infarction.

Abstract:Objective: To investigate the effect of astragalus saponins on mice myocarditis caused by CVB3. Methods: Male BALB/c mice was inoculated i.p. with 1000 TCID50 CVB3 and administered 5.64× 10-3 mg/g astragalus saponins i.p. for 7d starting 1 h after viral inoculation. The survial rate,the body temperature, LDH and GOT,the heart histopathologic changes,the virus titer of heart were observed. Results: The survival rate was higher, LDH and GOT release was lower, the body temperature was higher and similar to normal mice, the virus titer of heart was lower, the heart mass/body mass ratio was lower, the histopathologic changes (necrosis, degeneration and cellular infiltration) was lighter in astragalus saponins treated group than in viral control group. The statistic is valuelar. Conclusion: Astragalus saponins has therapeutic effects on viral myocarditis. The machenism may be anti-viral, improving of circulation and lightening of myocardial injury

Abstract:Objective:To investigate the expression of vascular endothelial growth factor (VEGF) in atherosclerotic plaques.Methods: The rabbit atherosclerotic model was developed by high cholesterol feeding.The VEGF protein level in atherosclerotic plaques were studied by means of immunohistochemistry.Results:VEGF protein was not detected in normal rabbit aortic arteries,the increased production of VEGF protein were found in atherosclerotic plaques.Conclusion:VEGF protein have a high level expression in atherosclerotic plaques and may involve the pathogenesis and progression of atherosclerosis.

Abstract:Objective:To investigate the immunosuppressive effects of leflunomide(Lef) in various organ transplantation models in rats. Methods: Sixty male Lewis rats received othortopic liver, small bowel, kidney and heterotopic heart, pancreas transplantation from ACI rats. Lef in its powder form was suspended in 1%carboxymethylcellulose and was administered at 5 mg.kg-1.d-1 by gavage for 14 d. Results: The survival time of rats in Lef group is significantly longer than the corresponding untreated control group.Besides, 2/6 rats with kidney transplantation and 5/5 rats with liver transplantation were alive over 100 d. Conclusion: Lef as monotherapy in short term treatment can delay acute rejection in heart, pancreas and small bowel transplantation, and lead to long term functional graft survival in kidney and liver transplantation in strong rejecting rat combination. In view of the effect of organ preference by Lef, this agent may be of special benefit to clinical kidney and liver transplantation.

Abstract:Objective: To evaluate the relation between DNA contents, 2c deviation indexes(2cDI) of colorectal cancer cells,changes of some gastrointestinal hormones levels,and the cancer clinical pathologic features. Methods: In 40 patients with colorectal cancer, DNA contents, 2cDI of the tumor cells (determined by automatical imaging cytometry), serum gastrin, plasma somatostatin(SS),tumor SS, cancer-adjacent mucosa SS and cancer-distant mucosa SS levels (assayed by RIA), and the cancer clinical pathologic features were analyzed by multiple variable stepwise regression with a computer. Results: The increases of tumor SS closely accompanied by the decrease of the cancer cell DNA contents or 2cDI (P<0.01). Cancer-adjacent mucosa SS had a significant positive correlation to the cancer cell DNA contents or 2cDIs (P<0.05 and P<0.01, respectively). The cancer cell DNA contents and 2cDIs increased significantly in patients with rectal cancer (P<0.01). Conclusion: The endogenous SS may play an important role in the regulation of DNA synthesis in cancer cell, elevating the tumor SS level and keeping the cancer-adjacent mucosa SS at high level, thus inhibiting the DNA sythesis in colorectal cancer cell.

Abstract:Objective:To assess the clinical significance of plasma neuropeptides in patients with viral hepatitis. Methods: Levels of plasma leucinenkephalin (LENK) and neuropeptide Y (NPY) were detected with radioimmunoassay in 67 patients with hepatitis and 10 healthy controls. Results: The level of LENK in patients was significantly higher than that in controls (P<0.05). In patients with hepatitis, the level of LENK was higher in chronic hepatic failure and chronic hepatitis (severe) than that in the rest (P<0.01). The concentration of LENK was positively correlative with serum total bilirubin (TBIL) (r=0.43, P<0.01), but negatively correlative with serum albumin (ALB) (r=-0.25, P<0.05) and prothrombin time activity (PA) (r=-0.55, P<0.01). The levels of NPY in chronic hepatic failure and chronic hepatitis (severe and medium) were significantly lower than that in other viral hepatitis groups and healthy controls (P<0.01). The concentration of plasma NPY in chronic hepatitis was negatively correlated with TBIL (r=-0.34, P<0.01), but postively correlated with ALB and PA (each r=0.38, P<0.01). Conclusion: Plasma LENK and NPY levels may be used as an associated marker for judging liver function. They may contribute to the pathogenesis of viral hepatitis and some of its symptoms.

Abstract:Objective:To study the rational therapeutic regimen in the treatment of severe acute pancreatitis (SAP). Methods:One hundred and fifty-eight cases of SAP were admitted to our department during the past 20 years (from 1980 to 1998),comparison was made between 73 cases in group A who were treated mainly by early operation and 85 cases in group B who were treated nonoperatively in the terms of complications rate, the mean hospitalization days and the hospitalization fee. Results:The incidence of complication, the mean hospitalization days and the hospitalization fee was lower in group B than in group A (P<0.01). Conclusion:It is an effective methods to adopt the combined nonoperative treatment in early SAP, and surgical operation is a rational addition.

Abstract:Objective: To study the nerve degeneration of lower extremity after paraplegia. Methods: (1) The spine cord of experimental dogs were severed between T11-12. The section of main nerve trunks of lower extremities were studied under microscope after 3-6 weeks; (2) the sural nerve of later paraplegic patients were analyzed. Results: No obvious degeneration was found in the nerve of lower extremities of experimental dogs and later paraplegic patients. Conclusion: No obvious degeneration is found in lower extremity nerve if the injury plane is higher than T11.

Abstract:Objective:To study the change of CTG repeat numbers in patients and their family members of DM, SEP and MEP.Methods: CTG repeat numbers of DM, SEP and MEP gene from 5 patients diagnosed as DM clinically, and their 16 family members and a healthy subject were analysed by polymerase chain reaction and Southern blot. Results: The numbers of CTG repeat were 19-30 in 10 normal individuals,SEP and MEP were normal. It ranged from 80 to more than 1 605 in 5 patients with DM which were much higher than the normal person. The repeat numbers of CTG in 12 of 16 family members were higher than the normal. The severity degree of clinical symptoms of these 5 patients was related to the degree of amplification of CTG, SEP and MEP.Conclusion: The gene diagnosis of DM is consistent with its clinical diagnosis, SEP and MEP.

Abstract:Objective:To evaluate the relationship between serum AGE and diabetic chronic complication (DCC). Methods: Serum AGE in diabetic patients was measured by competitive ELISA. Results: Both of 15 diabetic patients with DCC and 11 diabetic patients without DCC had higher mean serum AGE concentrations than that of normal group (P<0.01). Conclusion: This may be the increasing production of endogenous AGE that the diabetic group had a higher mean serum AGE than normal group,serum AGE may provide an indicators of long term blood glucose control and predict the occurances of diabetic chronic complications.
