    • Eukaryotic expression of human ICOS-Ig fusion protein and analysis of its biological activity

      Abstract:Objective:To express human ICOS extracellular region and IgG Fc fusion protein using euckaryotic vector and to analyze its biological activity.Methods: Human ICOS extracellular region and IgG Fc fragment were cloned by RT-PCR and inserted into eukaryotic expression vector pSecTag2.The constructed plasmid pSecTag2-ICOS-Ig was stably transfected into CHO cells.Soluble ICOS-Ig fusion protein was collected from serum-free medium and purified with protein A affinity chromatography.The purified product was analyzed by SDS-PAGE,Western blotting assay, and ELISA.Fluorescence-activated cell sorting (FACS) and mixed lymphocyte reaction (MLR) were used to study the activity of the fusion protein.Results: ICOS extracellular region and IgG Fc fragment were successfully cloned into expression vector; ICOS-Ig fusion protein was expressed and purified in mammal cells.The purified fusion protein specifically bound to ICOSL and inhibited mixed lymphocyte reaction.Conclusion: A ICOS-Ig fusion protein expression system has been successfully constructed,and bioactive ICOS-Ig fusion protein has been obtained.

    • Expression of angiotensin-converting enzyme 2 is increased during preservation injury of liver transplantation in rats

      Abstract:Objective:To observe the expression of local angiotensin-converting enzyme 2 (ACE-2) in rat liver and its relationship with preservation injury (PI) after transplantation,so as to determine the possible role of local renin-angiotensin system (RAS) in PI caused by the cold-preservation in liver transplantation.Methods:The donor liver grafts were divided into cold preservation (CP) and non-cold preservation (NCP) groups;we also included a sham operation group without transplantation.The mRNA and protein levels of ACE-2 in the transplanted livers were detected by quantitative real-time PCR and Western blotting assay,respectively.The location of ACE-2 protein and pimonidazole in liver was detected by immunohistochemistry.Furthermore,histological evaluation was applied to verify the extent of liver damage.Results:Histological injuries of different degrees existed after liver transplantation; the injury was more obvious in the CP group.Compared with the sham operation group,the mRNA and protein expression of ACE-2 was significantly increased after liver transplantation (P<0.05 or P<0.01),and the expression of the CP group was significantly higher than that of the NCP group (P<0.05).Pimonidazole staining was positive around the hepatic veins in the transplantation groups,and the intensity of staining was positively correlated with the expression of ACE-2 mRNA (r=0.78,P<0.001).Conclusion:It is indicated that ACE-2 expression is closely associated with tissue hypoxia during cold preservation-induced PI ,and local RAS may play an important role in PI during liver transplantation.

      Abstract:Objective:To establish a peptide nucleic acidmediated onestep PCR assay for detecting Kras mutation,and to evaluate its diagnostic value.Methods: We developed a onestep polymerase chain reaction (PCR) approach with melting curve analysis using wildtype specific peptide nucleic acid (PNA) and fluorescent dye SYBR Green Ⅰ to determine the genotypes in codon 12 and 13 of Kras oncogene. The result of our method was compared with that of restriction fragment length polymorphism (RFLP) analysis.Results: Our method simultaneously examined condon 12 and 13 of Kras oncogene,and was easy to perform.The sensitivity of our method was 0.001% when in a 105fold excess of wildtype Kras DNA.The detection limit was 102 copies.Our method could be used for large sample test,with the time of a single test being 1 hour.The limit of traditional method was 103copies and the sensitivity was 0.01%.The testing time was about 2 days for samples which needed only 1 hour using our method.Conclusion: Our method has obvious advantage over RFLP analysis; it is suitable for detecting Kras mutation in large samples and can be used as an effective method for early diagnosis of pancreatic cancer.

      Abstract:Objective:To explore the activity of Nacetylglucosaminyltransferase Ⅴ(GnTⅤ) in breast cancer tissues and to discuss its clinical significance.Methods:The activities of GnTⅤwere examined in a tissue microarray cohort of 57 breast carcinomas and 12 benign breast lesions.Lectin blotting using Phaseolus vulgaris leukocytic phytohemagglutinin (PHAL) labeling was used to assay GnTⅤactivity.The relationship between clinicopathological parameters and GnTⅤactivity was investigated.Results:High activity of GnTⅤwas observed in 89.47% (51/57) breast cancer tissues,which was significantly higher than that in the benign breast lesions (8.33%\[1/12\],P<0.01).No difference in GnTⅤactivities was observed in breast cancer patients with different pathological types,different lymphatic statuses,and different stages.Conclusion:Higher GnTⅤactivity in breast cancer tissues indicates a higher risk of malignancy.Targeting GnTⅤmight be a future way for treatment of triplenegative breast cancer patients with recurrence and metastasis.

      Abstract:Objective:To investigate the expression of cyclooxygenase2 (COX2),urokinase plasminogen activator(uPA),and thrombospondin1 (TSP1) in gastric carcinoma and the surrounding lymph nodes,so as to discuss their roles in the invasion and metastasis of gastric carcinoma.Methods:The expression of COX2,uPA,and TSP1 in the samples was examined by tissue microarray and SP immunohistochemistry staining.Results:The positive rates of COX2,uPA,and TSP1 in gastric carcinoma were 67.7%,78.1%,and 78.6%,respectively,which were significantly higher than those in the surrounding lymph nodes(40.0%,6.7%, and 40.0%,respectively,P<0.05).The expression of COX2 was significantly correlated with the depth of invasion (P<0.05).The expression of uPA was significantly correlated with lymph node metastasis and depth of invasion (P<0.05).The expression of TSP1 was neither correlated with lymph node metastasis nor depth of invasion (P>0.05).COX2,uPA,and TSP1 expression was not significantly different between metastatic lymph nodes and corresponding gastric carcinoma.COX2 expression was positively correlated with uPA,and TSP1 expression.TSP1 expression was not correlated with uPA expression.Conclusion:COX2,uPA,and TSP1 are highly expressed in the gastric carcinoma.COX2 expression is correlated with depth of invasion.UPA expression is significantly related with depth of invasion and lymph node metastasis.

    • Expression of EGFR,Grb2,p-mTOR and VEGF in human colorectal cancer tissues

      Abstract:Objective:To observe the expression of phosphorylated mammalian target of rapamycin(p-mTOR),epidermal growth factor receptor(EGFR),growth factor receptor-bound protein 2(Grb2) and vascular endothelial growth factor(VEGF) in human colorectal cancer tissues and to explore their roles in the carcinogenesis of colorectal cancer.Methods: Tissue microarray containing 185 colorectal cancer tissues was constructed and the expression of EGFR,Grb2,p-mTOR and VEGF in the colorectal cancer tissues and the corresponding adjacent tissues was examined by immunohistochemistry methods.The relationship between their expression with the clinicopathological characteristics such as age,sex,invasion depth,lymphatic metastasis,clinical stage and differentiation degree was analyzed.Results: EGFR,Grb2,p-mTOR and VEGF were scarcely expressed or absent in the corresponding adjacent tissues; their positive rates in the colorectal caner tissues were 21.1%,44.9%,42.2% and 54.1%,respectively,which were significantly higher than those in the corresponding adjacent tissues(P<0.05). The expression of EGFR,Grb2,p-mTOR and VEGF was not correlated with the patients’ sex,age and differentiation degree of cancer.Over-expression of EGFR was found significantly associated with the invasion depth and clinical stage of cancer(P<0.05); and over-expression of p-mTOR and VEGF was significantly associated with lymphatic metastasis,invasion depth and clinical stage(P<0.05).There was a correlation between every two of the four proteins (r=0.245-0.567,P<0.05).Conclusion: Over-expression of EGFR,Grb2,p-mTOR and VEGF is closely associated with the development and progresssion of colorectal cancer,and they may be worth further studying as new targets for the molecular target therapy of colorectal cancer.

      Abstract:Objective:To investigate the expression of PIAS2 in the testis tissues of patients with different spermatogenic functions.Methods:Eighty testicular biopsy samples from patients with azoospermia were subjected to pathological diagnosis and were divided into the following groups according to the pathological findings:normal or basically normal spermatogenesis group (n=40),hypo-spermatogenesis group (HS,n=20),and sertoli cell-only syndrome group (SCOS,n=20).Immunohistochemical method (SP) was used to examine the expression of PIAS2 in the testis tissues with different spermatogenic functions.Results:PIAS2 was strongly positive (dark brown) in the spermatogenic cells of different functions.It was weakly positive or absent in the Leydig cells and Sertoli cells.Conclusion:The status of PIAS2 in the testicular tissues is associated with the spermatogenic functions,and it may serve as a potential parameter for predicting of spermatogenic function.

    • Selection of peptides bind to PreS1 antigen from a phage-displayed peptide library

      Abstract:Objective:To screen for peptides that specifically bind to PreS1 antigen from a phage-displayed peptide library.Methods: The PreS1 antigen was used as the target molecule to screen the binding peptide from the Ph.D.-7 peptide library with phage display technique,and the positive clones were identified by ELISA.Results: After three rounds of biopanning,the binding peptides were screened from the peptide library by ELISA and competitive inhibiting ELISA.Sequencing result showed that the binding peptides had high affinity and specificity.Conclusion: A peptide binding PreS1 antigen has been successfully obtained by screening the phage display library,which paves a way for the diagnosis and treatment of hepatitis B infection.

    • Anti-aging activity of Mytilus coruscus polysaccharide in aged SD rats

      Abstract:Objective:To investigate the anti-aging activity of Mytilus coruscus polysaccharide (MP) in aged rats.Methods:We established a 24-month natural aging rat model.Twenty model rats were divided into 4 groups,namely,the aging model group,Mytilus coruscus powder group (MCP 1 g/kg),high dose MP group (MP 400 mg/kg),and low dose MP group (MP 200 mg/kg).Five three-month-old rats served as young control.The Mytilus coruscus power solutions and MP solutions were given intragastrically on a daily basis for 90 days.The organ indices of the heart,liver,kidney,and spleen of rats were observed; the pathology of the hearts and livers was also observed.The levels of superoxidedismutase (SOD),glutatio-neperoxidase (GSH-Px),and malonaldehyde(MDA) in the serum and liver were determined; and the main serum parameters were also determined.Results:Compared with the aging model group,the heart and liver pathology had no obvious aging changes in the high dose MP group.The serum and liver SOD,GSH-Px levels were significantly higher in the high dose MP group than those in the aging model group (P<0.01); the MDA level was significantly lower than those in the aging model group (P<0.01).Conclusion:The Mytilus coruscus polysaccharide possess anti-aging activity,and the anitoxidative effect might be one of the mechanism for the anti-aging effect.

    • Effects of tibolone on apoptosis of neurons after cerebral ischemia-reperfusion injury in rats

      Abstract:Objective:To study whether the neuroprotective effect of tibolone during cerebral ischemia injury is associated with its anti-apoptosis effect.Methods: A total of 30 healthy adult female Sprague Dawley rats were evenly randomized into sham-operated group,ovariectomized group,and tibolone group(0.25 mg/\[kg·d\]).Brain ischemia model was induced by left middle cerebral artery occlusion(MCAO) for 2 hours followed by 24 hours reperfusion.All animals were killed and the cerebral infarction volumes were determined by 2,3,5-triphenyltetrazlium chloride (TTC) staining and image analysis.Apoptotic cells were labeled by Terminal deoxynucleotidyl Transferase-mediated dUTP nick end labeling (TUNEL) .The expression of Bcl-2,Bax,and caspase-3 was analyzed by immunohistochemical SP method. Results: Compared with the sham-operated group(P<0.05),cerebral infarction volume was significantly reduced,TUNEL positive cells was significantly decreased(P<0.01),Bcl-2 positive cells were significantly increased,and Bax(P<0.01),caspase-3 postive cells were significantly reduced compared with tibolone treated groups (P<0.01).Conclusion: Tibolone can inhibit the neuron apoptosis following cerebral ischemia injury in OVX rats,displaying a neuroprotective effect.

    • Inhibitory effect of germanium against local expression of 5-HT and HA in arthritic rats

      Abstract:Objective:To investigate the effect of germanium against local expression of 5-hydroxytryptamine (5-HT) and histamine (HA) in adjuvant-induced arthritic rats.Methods: Male SD rats were randomly divided into 6 groups (n=10): blank control group,model group,low(10 mg/kg),medium(20 mg/kg),and high dose(40 mg/kg)β-carboxyethylgermanium sesquioxide (Ge-132) groups,and indomethacin(IDMT,1 mg/kg)control group.Rat model of rheumatoid arthritis (RA) was induced in the later 4 groups by Freund’s complete adjuvant (FCA).The body weights and the paw volumes of rats in all groups were determined before FCA injection and d 3,6,9,12,15,18,and 21 after injection; the rates of weight gain and paw swelling were calculated.The contents of HA and 5-HT in inflammatory tissues were measured at d 21 immediately after rats were beheaded.Results: The rates of weight gains in middle and high dose germanium groups and IDMT group were significantly higher than those in the low dose germanium group and model groups(P<0.01)at d 21,but had no difference with those of blank control group.The rates of paw swelling in the three germanium groups were significantly lower than that of the model group(P<0.01),and higher than that of the IDMT group(P<0.01 or 0.05) ; that of the high germanium group was significantly lower than those of the other 2 germanium groups (P< 0.05).The contents of HA in the local asthmatic tissues in the 3 germanium and IDMT groups were significantly lower than that of the model group,and those of the low and middle germanium groups were significantly higher than those of the higher germanium and IDMT groups (P<0.01).The contents of 5-HT in middle and high germanium and IDMT groups were significantly lower than those of the model group and low germanium(P<0.01).Conclusion: Middle and high dose germanium may relieve local asthmatic inflammation in rats through inhibiting the release of HA and 5-HT,with its effect similar to that of IDMT.

    • Influence of sodium selenite on adiponectin expression in kidney of rats with diabetic nephropathy

      Abstract:Objective:To investigate the influence of sodium selenite on adiponectin expression in kidney of rats with diabetic nephropathy and the relation between sodium selenite and adiponectin,so as to investigate the role of sodium selenite and adiponectin in diabetic nephropathy.Methods: Diabetic nephropathy rat models were established by injecting streptozocin intraperitoneally and high fat diet.The study was divided into 3 groups,namely,a control group,a diabetic nephropathy group and a selenite-treated group.Animals in the selenite-treated group were given selenite solution 1 μg/kg daily via intragastric administration; those in the other 2 groups were given the same dose of saline in the same way.The animals were sacrificed 10 weeks later and the blood and urine samples were used for biochemical examination.Samples of renal cortex were cut quickly and fixed with 4% glutaral for observation under scanning electron microscope.The renal samples were made into paraffin section for observation of the pathological changes under light microscope and for location of protein expression via immunohistochemistry analysis.The expression of adiponectin mRNA and protein was examined by real-time PCR and Western blotting,respectively.Results: The general condition and biochemical parameters of animals in the selenite-treated group were obviously improved and the pathological changes were obviously releived compared with those in the diabetic nephropathy group.Immunohistochemistry analysis showed that the expression of adiponection in the selenite-treated group was obviously stronger than that in the diabetic nephropathy group,and the expression was found in both glomcrulus and renal tubule.The expression of adiponectin mRNA in the selenite-treated group and diabetic nephropathy group was 2.043 and 1.373 folds that of the control group,respectively.The expression of adiponectin protein in the selenite-treated group was significantly higher than those in the control group and diabetic nephropathy group (P<0.05).Conclusion: Sodium selenite can enhance renal expression of adiponectin through anti-oxidative stress and promotion of periphery tissue sensitivity to insulin,indicating that sodium selenite and adiponectin may play important roles in delaying the progression of diabetic nephropathy.

    • Application of Markov model in studying graded prognosis of chronic kidney disease

      Abstract:Objective:To evaluate the progression of chronic kidney disease (CKD) in CKD patients and to establish a Markov model for graded prognosis of CKD.Methods:A total of 272 CKD patients were retrospectively investigated.A Markov model consisting of six states (CKD1 stage,CKD2 stage,CKD3 stage,CKD4 stage,CKD5 stage as well as death/ end-stage renal disease \[ESRD\] stage) was established.Results:The mean follow-up period was 2.0 years.Transition rates from CKD1 stage to CKD2 stage,from CKD2 stage to CKD3 stage,from CKD3 stage to CKD4 stage,from CKD4 stage to CKD5 stage and from CKD5 stage to death/ESRD stage were 9.2%/year,10.9%/year,13.2%/year,16.1%/year,and 47.1%/year,respectively.The Markov model estimated that the mean duration of CKD1 stage,CKD2 stage,CKD3 stage,CKD4 stage,CKD5 stage and death/ ESRD stage in our cohort were 11.1 years,7.8 years,5.4 years,2.5 years and 1.0 years,respectively.The mean renal survival time or dialysis free period was 27.8 years.Conclusion:Evaluation of severity and the treatment of CKD patients should be done according to the prognoses of CKD patients at different stages.

    • Interaction between ciprofloxacin and ferric-ion binding protein from pathogenic bacteria

      Abstract:Objective:To investigate the interaction between ciprofloxacin (CIP) and ferric-ion binding protein (FBP) from pathogenic bacteria,so as to determine whether FBP is the target of CIP.Methods: Reactions between ciprofloxacin and FBP from N.gonorrhoeae,which was expressed in E.coli,were monitored by UV-visible and NMR spectroscopy.Results: Fe3+ was removed from holo-FBP by CIP in 10 mmol/L Tris-Cl buffer,pH 7.40,at 298 K,and half of the loaded Fe3+ was taken off when 50 molar equivalence of CIP was present.Conclusion: CIP can remove Fe3+ from holo-FBP,indicating that CIP can interfere with the Fe3+ capture of the bacteria and influence the growth and virulence of the bacteria.

      Abstract:Objective:To identify the chemical components in traditional Chinese herbal medicine Ginseng by high performance liquid chromatography diode array detector (HPLC-DAD) and High performance liquid chromatography-time of flight mass spectrometry (HPLC-TOF/MS).Methods: An Agilent Zorbax XDB-Cl8 column (250 mm×4.6 mm,5 μm) was used for isolation and identification of chemical components in Ginseng with a mobile phase of acetonitrile and water in gradient elution.A gradient program was used as follows:1-35 min,19% A; 35-55 min,19%-29% A; 55-70 min,29% A; 70-110 min,29%-40% A; 110-150 min,95% A.The flow rate was set at 1.0 ml/min,the injection volume was 20 μl,and the column temperature was set at 20℃.The time-of-flight mass spectrometer was equipped with an EIS ion source.Scanning mass range was between m/z 100-1 350.Results: Thirty-nine chemical compounds in Ginseng were identified unequivocally.Conclusion: Chromatographic demonstration of 39 chemical compounds of Ginseng in one run is achieved by HPLC-TOF/MS,which provides a foundation for further study on the metabolism and action mechanism of traditional Chinese herbal medicine.

      Abstract:Objective:To analyze the chemical constituents of Rhizoma belamcandae by using high-performance liquid chromatography-diode array detector/electrospray ionization-mass spectrometry (HPLC-DAD/ESI-MS).Methods: Dried Belamcanda chinensis powder was extracted with 70% ethanol by sonication.The chromatographic separation was performed on a YMC ODS-C18 column (250 mm×4.6 mm I.D.,5 μm) with a mobile phase composed of 20%CH3OH(A)-70%ACN(B) (0→30→45→65 min,20→30→90→100 B),eluted at a flow rate of 0.45 ml/min,and the UV detection wavelength was set at 265 nm.Positive ionization mode with a needle voltage of 5 000 V,a capillary voltage of 20 V,a gas(N2)press of 20 psi and a temperature of the drying gas of 300℃ was selected.Relative molecular mass data acquisition was performed from m/z 250 to 550 in full MS scan mode.Results: Nine major isoflavones were identified from Rhizoma belamcandae based on their retention behavior obtained on-line by their UV spectra and the HPLC-DAD/ESI-MS.Conclusion: A rapid and efficient HPLC-DAD/ESI-MS method for identifying the chemical constituents of Belamcandae chinensis has been established,which provides more scientific information for quality control of Rhizoma belamcandae.

    • Synthesis of pyridazinone derivatives and study of their antiplatelet aggregation activity

      Abstract:Objective:To study the influence of different amino group introductions on the antiplatelet aggregation activities of pyridazinone derivatives.Methods: The target compounds were designed and synthesized by inletting different substitution amino groups using acetyl fragment as the connecting segment.Born method was used for preliminary pharmacological test in vitro.Results: Ten target compounds were designed and synthesized; 8 of them were reported firstly and all of them were confirmed by 1HNMR spectra.The results of preliminary pharmacological test showed that all the target compounds exhibited potent antiplatelet aggregation activity.The antiaggregation activities of compounds (6f),(6g),(6h) and (6j) were stong; compounds (6g) and (6j) showed a 5-time higher activity than 6-\[4-(pyridin-4-yl-amino)phenyl\]-4,5-dihydro-3(2H)-pyridazinone (MCI-154).Conclusion: Inletting different substitution amino groups can enhance the antiplatelet aggregation activity of the compounds.

    • Establishment of hsa-mir-122 stably transfected-HepG2 cell line and its lipid metabolism characteristics

      Abstract:Objective:To establish a HepG2 cell line stably overexpressing hsa-mir-122 by transfecting the HepG2 cells with pEZX-hsa-mir-122 vectors,so as to observe the influence of miR-122 on the lipid metabolism in the liver cells.Methods:The constructed pEZX-hsa-mir-122 and pEZX-mir-control plasmids were identified by restriction enzyme digestion and the right clones by sequencing were subjected to sequencing.And the right clones by sequencing were transfected by LipofectamineTM.The transfection efficiency was assessed by green fluorescent protein expression.The stably transfected cell lines were screened by Puromycin.The expression of miR-122 was examined by real-time PCR.The inhibitory function of miR-122 was testified by Western blotting assay.Nile red was used to observe the content of lipid in HepG2 cells stably overexpressing hsa-mir-122.Results:HepG2 cells were successfully transfected with pEZX-hsa-mir-122 and pEZX-mir-control plasmids.The precursor miRNA sequences were integrated into the genome of HepG2 cells stably overexpressing miR-122.Compared with the control HepG2 cells,the HepG2 cells overexpressing miR-122 contained more lipid in the cytoplasm.Conclusion:The miR-122 precursor sequence can intergrate into the genome of HepG2 cells.The increase of miR-122 level in HepG2 cells can result in abnormal metabolism of lipid.

      Abstract:Transient receptor potential vanilloid 1 (TRPV1) is a non-selective cation channel,which can be activated by multiple pathways during the course of the diseases.Recent studies indicate that primary sensory neurons of the pancreas express TRPV1 receptor and the activation of TRPV1 receptor promotes pancreatic inflammation.Moreover,blockade of these transient receptor potential channels can greatly ameliorate the pain response in experimental pancreatitis.

    • Cubosomes as drug delivery system: recent advance

      Abstract:Cubosomes are self-assembled nanostructured particles formed by aqueous lipid and surfactant systems.Cubosomes are thermodynamically stable; they have a structure like “honeycombed” with bicontinuous domains of water and lipid in which surfactant assembles into bilayers and twisted into a three dimension,periodic,and minimal surface,forming a tightly packed structure.The properties of cubosomes,such as its unique bicontinuous cubic phase liquid crystals,its ability to solubilize hydrophobic,hydrophilic,and amphiphilic molecules at the same time,its biodegradability by simple enzyme action,its strong bioadhesion ability,and its simple preparation,make them a promising vehicle for drug delivery.Based on recent reports,this review introduces the structure,preparation,exosyndrome and drug delivery potential of cubosomes.

      Abstract:Side population (SP) cells are highly enriched for stem cell activity and characterized by their ability to efflux the vital dye Hoechst 33342,because they express the ATP binding cassette (ABC)-dependent transporter ABCG2.SP cells can be selected from main population using flow cytometric analysis.Currently SP cells have been isolated from many tissues and organs.SP cells of different origins have some common characteristics.This article introduces the classifications,surface marker,and characteristics of SP cells.

      Abstract:Bone morphogenetic proteins (BMPs) play a pivotal role in the formation,induction and repair of the bone; BMPs have been confirmed to induce bone formation.Transforming growth factor-beta,insulin-like growth factor-Ⅰ and basic fibroblast growth factor also contribute to bone formation; combination of the factors with BMPs can promote the healing of fracture and non-union,which may have a bright future in orthopedics.

    • Determination method for in vitro disintegrating time of oral disintegrating tablets of Chinese herbal drug

      Abstract:Objective:To develop methods for determining the disintegration time of oral disintegrating tablets (ODTs) of Chinese herbal drug,and to select the evaluation method with good reproducibility,resolving power,and in vivo and in vitro correlation.Methods: Several placebo ODTs and Chinese herbal drug ODTs were prepared.Their in vitro disintegration time was determined by self-developed method and routine methods,and the results were compared with the oral disintegration time.The reproducibility,resolving power,and the correlation between in vivo and in vitro of those methods were evaluated.Results: Different determination methods obtained different disintegration results of the same tablet.Graduated cylinder and improved disintegration apparatus both had shortcomings and their determination results were very different from that of the oral disintegration time.Our self-developed disintegration device could better determine the disintegration ability of the ODTs,and the result was closer to the oral disintegration time,with clear end point of disintegration and better reproducibility,resolving power,and the in vivo and in vitro correlation compared with other methods.Conclusion: Our self-developed device is suitable to determine in vitro disintegration time of ODTs,and may serve as a reference for quality control of ODTs.

    • Development of quantitative ELISA for determination of serum adiponectin and its clinical application

      Abstract:目的:建立一种检测人血清中脂联素(APN)的酶联免疫吸附方法(ELISA),并初步探讨其在冠心病诊断中的应用。方法:构建表达质粒pET22b(+)/gAPN,在原核表达系统大肠杆菌中表达并纯化重组脂联素球状区(gAPN)蛋白。用抗APN单克隆抗体MAB10651包被微孔板,10%小牛血清作为封闭液,针对APN不同抗原表位的单克隆抗体MAB1065生物素化后作为标记抗体,联合生物素-亲和素信号放大系统,以gAPN重组蛋白为标准品绘制标准曲线,以准确性、特异性、重复性、回收率试验和方法对比试验评价ELISA方法。检测161例冠心病患者血清APN水平并结合冠状动脉造影结果和Gensini积分系统,探讨血清APN水平与冠心病的关系。结果:利用基因克隆技术成功构建了pET22b(+)/gAPN质粒,获得了相对分子质量约15 000的重组gAPN蛋白,产量较高(1.57 mg),纯度>90%,并以此为标准品初步建立了检测人血清APN的定量ELISA方法。该法具有良好的准确性和重复性,灵敏度达150 pg/ml,回收率为91.0%~108.0%,与深圳依诺金生物科技有限公司进口分装的ELISA试剂盒对照比较有良好的相关性(r=0.935,P<0.001)。临床检测表明冠心病组血清APN水平明显低于非冠心病组(P<0.01),且血清APN水平随冠状动脉狭窄程度的加重呈进行性下降趋势。结论:本实验成功建立了一种简便快速检测人血清APN的ELISA方法,对诊断冠心病有临床应用价值,为国内APN诊断试剂盒的研制和开发提供了科学实验依据。

      Abstract:目的:采用Meta分析的方法探讨健康教育对糖尿病足的疗效影响。 方法:检索主要医学数据库以获取相关文献,按一定纳入和排除标准筛选文献,共纳入9篇随机对照试验研究文献,采用Review Manager V 5.0软件从自我护理能力、治疗有效率、生活质量、住院时间、空腹血糖值、知识掌握6个方面进行Meta分析。 结果:健康教育组与对照组在自我护理能力、生活质量、住院时间、空腹血糖值、知识掌握方面之间的差异具有显著性意义(P<0.01),而在治疗有效率方面之间的差异无显著性意义。 结论:目前医护人员在常规治疗以外针对糖尿病足患者及其家属进行的健康教育,在一定程度上改善了患者的治疗效果。但由于诸多因素,健康教育的效果并不理想,应对糖尿病足的健康教育作进一步研究和探讨。

      Abstract:食管穿孔与破裂是一种较严重的疾病,发病率虽然较低,但病情发展很迅速。食管穿孔、破裂后消化液及细菌自食管破孔迅速外溢,可引起纵隔或胸膜腔急性化脓性炎症,如不能及时诊断和正确治疗病死率较高,可达20%以上[1-2]。及时诊断和正确治疗是抢救患者生命的关键。现总结我院2006~2008年来24例行CT及X线食管造影检查的食管穿孔与破裂病例,以加深对本病的认识。

      Abstract:高血压病是老年人群的常见疾病,与动脉粥样硬化的发生密切相关。颈动脉是全身中型动脉的代表,其硬化程度可间接反映冠状动脉、外周动脉等的硬化程度,可作为一个良好的窗口来监测体内动脉粥样硬化的进展,有效评估高血压患者靶器官损伤程度\[1-2\]。动态血压监测(ambulatory blood pressure monitoring, ABPM)可如实反映患者血压的波动和昼夜节律变化等,对高血压病的诊治及预后判断具有较大意义\[3-4\]。因此,本研究联合应用颈动脉超声结合ABPM技术探讨老年高血压病患者的昼夜节律变化与颈动脉粥样硬化斑块的相关性,为老年高血压病患者靶器官损害的防治提供依据。

      Abstract:气道内出血是呼吸科常见的危急病症之一,通常需要紧急实施纤维支气管镜气道内出血止血术\[1-3\]。目前此手术多采用保留自主呼吸的静脉全麻,但其可导致呼吸中枢抑制、喉痉挛、术中体动、呛咳和屏气等并发症,术中风险较高\[1\]。国内不少学者\[2-3\]近年来尝试将面罩、内镜面罩通气应用于气道内镜手术,在呼吸保障及控制方面取得较好效果。因此,本研究进一步尝试在全麻纤维支气管镜气道内出血止血术中应用三通喉罩(three-way laryngeal mask airway,TLMA)通气,探讨其临床效果,并评价其安全性。

      Abstract:近年来的研究\[1-2\]证实,三氧化二砷(As2O3)不仅对急性早幼粒细胞白血病有良好的疗效,而且在实体肿瘤的治疗中也显示了良好的前景。目前对As2O3治疗恶性胸腹腔积液的机制研究不够深入,临床上多为个案报道。本研究应用As2O3胸腔内灌注治疗肺癌合并恶性胸腔积液,通过与博莱霉素(BLM)做平行对照,评价As2O3用于治疗肺癌合并胸腔积液的疗效、安全性及对患者耐受性的影响,以探讨其在临床应用中的价值。

      Abstract:1.1一般资料患者,女,74岁,因“反复胸闷、胸痛2年,加重1周”于2007年9月7日入院。查体:神志清楚,血压110/60 mmHg(1 mmHg=0.133 kPa),心率78次/min,律齐,无异常杂音。心电图示窦性心律,急性广泛前壁及下壁心肌梗死。急诊冠状动脉造影显示左前降支开口处狭窄75%,左回旋支近段狭窄90%,右冠状动脉开口处狭窄90%。完善相关准备后,于左回旋支近段病变处放置药物洗脱支架1枚,术后给予低分子肝素5 000单位(每天2次,10 d),以及阿司匹林、氯吡格雷等药物治疗。
