Abstract:Objective To investigate the pathological roles of postnatally up-regulated alpha-fetoprotein(AFP) expression in ZBTB20 knockout mice in postnatal growth retardation, metabolic dysfunction, and pre-mature mortality. Methods ZBTB20 and AFP knockout mice were generated by crossbreeding ZBTB20 knockout mice with AFP knockout mice.The postnatal growth, survival rate, and blood glucose of ZBTB20/AFP double knockout mice were observed and compared with those of ZBTB20 knockout mice. Results There were no significant differences in postnatal growth, survival, and glucose homeostasis between ZBTB20/AFP doube knockout mice and ZBTB20 knockout mice. Conclusion The postnatally up-regulated liver AFP in ZBTB20 knockout mice is not associated with the growth retardation, hypoglycemia, or pre-mature mortality.

Abstract:Objective To construct the mutants of oncogene p28GANK with different ankyrin repeats deleted, so as to further study the potential role of p28GANK in hepatocellular carcinoma (HCC).Methods QuikChang Site-Directed Mutagenesis Kit was used to construct five p28GANK mutants with different ankyrin repeats deleted and their influences on p53 were also investigated. Results Five deletion mutants of p28GANK were as follows: 39-71aa, 72-104aa, 105-137aa, 138-170aa, and 171-203aa. Agarose gel electrophoresis and Western blotting analysis confirmed the correct construction of these mutants. Wild type p28GANK promoted degradation of p53, but the 5 mutants had no noticeable effects on expression of p53.Conclusion We have successfully constructed five p28GANK ankyrin repeat-deleted mutants. It is confirmed that the regulation of p53 needs the complete ankyrin repeats of p28GANK, which paves a way for further research on ankyrin and the role of p28GANK in HCC.

Abstract:Objective To observe the morphological changes of the retina at early stage of streptozotocin (STZ)-induced diabetes mellitus (DM) model, so as to assess the feasibility of using STZ-induced DM as an animal model of early diabetic retinopathy (DR). Methods Sixty-four male SD rats (body weight \[180±20\] g) were randomly divided into the CON and DM groups (n=32). Rats in DM group received intraperitoneal injection of 1% STZ once and those in the CON group were given same volume of citrate buffer in the same manner. The average body weight and blood glucose were similar in the two groups before intraperitoneal injection. The blood glucose, body weight, and other parameters were determined once a week after intraperitoneal injection. In the 10th week, the morphological changes of the retina were observed by H-E staining, stretched preparation of FITC-dextran perfused retinal blood vessels, and transmission electron microscopy in randomly chosen samples. Results The successful rate of STZ-induced DM model was 100%. The body weight of animals in DM group had no obvious increase after injection, and it even decreased at late stage. The body weight increased gradually in the CON group. The average body weight of DM group (\[169.9±26.9\] g) was significantly lower than that of the CON group (\[439.2±23.5\] g) in the 10th week (P<0.001). The average blood glucose of DM group (\[26.63±4.54\] mmol/L) was significantly higher than that of the CON group (\[6.37±0.49\] mmol/L) 72 h after injection (P<0.001). DM group had a blood glucose > 16.7 mmol/L and CON group had a blood glucose of 5.6-7.4 mmol/L throughout the 10 weeks. Retina H-E staining showed retinal capillary dilatation and interstitial edema in DM group in the 10th week, and the CON group had no obvious abnormalities. Stretched preparation of FITC-Dextran perfused retinal blood vessels showed vascular tortuosity and caliber irregularity in the DM group, but with no leakage, microvascular tumor or retinal non-perfusion area. The retinal vessel diameter was uniform and the branching was natural and smooth in the CON group. TEM results showed the following retinal ultrastructural changes in the DM group: thicker capillary basement membrane, digitation of capillary endothelial cells, mitochondrial swelling, cristae disruption, and vacuolar degeneration in capillary endothelial cells, bipolar cells and ganglion cells, mitochondrial swelling and cristae disruption in pericytes, decreased membranous disc (photoreceptor cell’s outer segment), and widened gap between membranous discs. Conclusion The retina morphological changes of early stage background diabetic retinopathy(BDR) can be found in the 10th week after STZ injection in rats, and STZ-induced DR model can be used as an early stage DR model; besides, the method is simple, economical, quick, with good reproducibility and high successful rate.

Abstract:Objective To evaluate the different multi-detector computed tomography features of benign and malignant pulmonary focal ground-glass opacity (fGGO) nodules. Methods The demographic data(sex and age), nodule location, size, and multi-detector CT (MDCT) features (shape, margin, interface, internal characteristics, and adjacent structures) of 82 fGGO patients were retrospectively analyzed. Differences between benign and malignant fGGO were analyzed by using chi-square test, Fisher exact test, and Mann-Whitney U test. Binary logistic regression analysis was also performed to draw a regression equation. Results The 82 fGGO patients included 61 with malignant and 21 with benign nodules. No statistical differences were found between benign and malignant fGGO nodules in terms of demographic data, nodule location, or nodule size. Binary logistic regression analysis showed that lobulation, interface, and pleural indentation were the important features of malignant nodules, with the corresponding odds ratio (OR) being 8.122, 3.139, and 9.076, respectively. The sensitivity, specificity, and accuracy of the obtained equation were 93.4%, 66.7%, and 86.6%, respectively. Conclusion An fGGO nodule with lobulation, well-defined but coarse interface, and pleural indentation in its MDCT image, is more likely to be malignant.

Abstract:Objective To evaluate the success rate and potency rate of functional and traditional end-to-side anastomosis for radial artery-cephalic vein arteriovenous fistula in hemodialysis patients, so as to summarize our experience and to improve the operation outcomes.Methods Totally 124 patients with chronic renal failure receiving radial artery-cephalic vein arteriovenous shunt for hemodialysis were randomly divided into 2 groups, namely, the functional end-side anastomosis and the traditional end-side anastomosis. The successful rate and year patency rate of the two methods were compared and analyzed.Results The mean operation time periods for functional and traditional end-side anastomosis were (20.4±5.6) min and (26.2±5.2) min, respectively(P<0.05) . The 1-month, 6-month, and 12-month patency rates were 93.6%, 87.3%, and 76.2%, in the functional end-side anastomosis group, and 95.1% (P>0.05), 82.0%(P>0.05) ,72.1%(P>0.05) in the traditional end-side anastomosis group, respectively, with no significant differences found in the three rates between the two groups.Conclusion The short-term and long-term patency rates are similar between hemodialysis patients undergoing functional and traditional end-to-side anastomosis for radial artery-cephalic vein arteriovenous fistula. The functional end-side anastomosis is more convenient and needs a shorter time, and it also makes it easy for a second surgical exploration.

Abstract:Objective To investigate the safety and efficacy of percutaneous microwave ablation (PMWA) for huge liver cavernous hemangiomas(LCHs), and to assess the potential advantages of extracting blood from the hemangioma during the ablation procedures. Methods A water-cooled microwave antenna named KY-2100 compatible with a frequency of 2,450 MHz generator was used to perform heating ablation in 19 patients for their LCHs guided by contrast-enhanced ultrasound under general anesthesia; the patients had multiple LCHs, at least with one having a diameter longer than 6 cm. Twenty lesions matched in their maximal size in 10 patients were ablated assisted with or without extracting blood from the hemangiomas. The relevant factors for the safety and ablation duration, thoroughness of ablation, and complications were comprehensively assessed. Results The ablation durations varied from 39 to 163 min in the 19 patients, with an average of (93±39.85) min. For those maximum-length matched lesions, blood extraction significantly shortened the ablation duration compared to non-blood extraction group (\[29.61±14.07\] min vs \[41.57±14.93\] min, P<0.05). Except for slight elevation of serum ALT level (\[133.58±46.29\] U/L) in 75% patients, there were no other complications such as intra-peritoneal bleeding, bile leakage, or hemoglobinemia. The average hospital stay was (4±0.95) days. All the ablated LCHs shrank markedly, with the maximal diameter decreased by 82.54%. The hemangioma-induced symptoms were relieved in all patients. Conclusion PMWA is technically feasible, safe and effective for treating LCHs. It can be an attractive alternative for surgical resection of hemangiomas at high-risk locations. And prompt extraction of focal blood can reduce the bleeding risk and shorten ablation duration.

Abstract:ObjectiveTo compare the clinical outcomes of intubation between HPHJ-A video laryngoscope and Macintosh laryngoscope, and to analyze the advantages and disadvantages of each approach.MethodsTotally 100 patients who were to undergo general anesthesia with endotracheal intubation were randomly allocated for airway management with one of the two devices: HPHJ-A video laryngoscope and Macintosh laryngoscope. The following data were observed and recorded for the two groups: the Cormack-Lehane grade of exposing the glottis, hemodynamic alteration before and after endotracheal intubation, intubation time, and intubation attempts.ResultsHPHJ-A video laryngoscope resulted in a better glottis exposure than the Macintosh laryngoscope, and the results of hemodynamic alteration, intubation time, and intubation attempts were similar between the two groups. For patients with a difficult airway (Cormack-Lehane Ⅲ-Ⅳ) , HPHJ-A video laryngoscope resulted in a slighter hemodynamic alteration(P<0.05), a higher successful intubation rate, and a shorter intubation time(P<0.05) compared with Macintosh laryngoscope. ConclusionThe HPHJ-A video laryngoscope can greatly improve the glottic exposure compared with the Macintosh laryngoscope, especially for patients with difficult airway.

Abstract:ObjectiveTo determine the serum HER-2/neu autoantibody level in patients with epithelial ovarian cancer and to evaluate the potential of using HER-2/neuautoantibody as a tumor marker. MethodsThe serum HER-2/neu autoantibody levels were measured by ELISA in 20 normal controls, 20 patients with benign epithelial ovarian tumors and 40 patients with epithelial ovarian cancer during 2007 to 2009. Statistical analysis was performed to determine the diagnostic value of serum HER-2/neu autoantibody. ResultsThe sera of epithelial ovarian cancer patients expressed higher levels of HER-2/neu autoantibody than those of benign epithelial ovarian tumor patients and normal controls (P<0.05). The ROC curve results showed that the HER-2/neu autoantibody screening exhibited a sensitivity of 47.5% and a specificity of 97.5% for diagnosis of epithelial ovarian cancer. Compared with serum CA125 measurement, the combined measurement of CA125 and HER-2/neu autoantibody yielded an increased specificity of 90% without decreasing the sensitivity. And no association was found for serum CA125 and HER-2/neu autoantibody using partial correlation coefficient. ConclusionIt is shown that the serum HER-2/neu autoantibody level is associated with epithelial ovarian cancer and can serve as a potential serological biomarker for epithelial ovarian cancer diagnosis.

Abstract:ObjectiveTo investigate the changes of TFF2 and TFF3 expression in sublingual gland during spontaneous healing of experimental gastric ulcer in rats. MethodsA total of 48 male SD rats were divided into gastric ulcer group (n=42, ulcers were induced by injection of acetic acid to the submucosal of paries anterior gastricus) and normal group (n=6). Immunohistochemical and RT-PCR methods were used to examine the expression of TFF2 and TFF3 in the sublingual gland in gastric ulcer group and normal group.ResultsImmunohistochemical staining showed that positive signals of TFF2 and TFF3 were mainly located in the striated duct, intercalated duct, myoepithelial cells, and some lumens. Compared with the control group, the integrated optical density (IOD) value of TFF2 was obviously increased on day 1 after gastric ulcer(P<0.01),and reached the bottom on day 2, then gradually increased again on day 4, 6 (P<0.01) , and kept at a high level during day 10-23 (P<0.05). The IOD value of TFF3 was similar to that of the normal group on day 1, 2 after gastric ulcer, then gradually increased on day 4, 6 (P<0.05), reached the peak on day 10 after gastric ulcer (P<0.01), and kept at a high level till day 23 after gastric ulcer(P<0.05). The results of RT-PCR showed that the changes of TFF2 mRNA and TFF3 mRNA were basically consistent with the change of their corresponding peptides. ConclusionTFF2 and TFF3 expression is increased in the sublingual gland during the spontaneous healing of experimental gastric ulcer in rats, and they may participate in the healing process.

Abstract:ObjectiveTo observe the effects of Tongluo recipe on the renal expression of connective tissue growth factor(CTGF), advanced glycation end products receptor(RAGE) and phosphorylated extracellular signal regulated kinase(p-ERK1/2) in streptozotocin-induced rat model of type 1 diabetes mellitus, and to explore the mechanism of its renoprotective effect. MethodsAdult male Sprague-Dawley rats, weighing 180-220 g, were randomly divided into 2 groups: control group (A, n=10) and model group (n=50). Rat diabetic model was induced by intraperitoneal injection of streptozotocin (60 mg/kg body weight), and the model rats were randomly subdivided into B group (diabetic control, n=10),C group(diabetic rats treated with low dose of Tongluo recipe,0.5 g·kg-1·d-1, n=10), D group (diabetic rats treated with medium dose of Tongluo recipe, 1.0 g·kg-1·d-1, n=10), E group (diabetic rats treated with high dose of Tongluo recipe, 2.0 g·kg-1·d-1, n=10), and F group (diabetic rats treated with losartan, 30 g·kg-1·d-1, n=10). At the end of the 12th week the rats were sacrificed and were weighed; the urea nitrogen (BUN) in blood, serum creatinine (Scr), 24 h albumin excretion rate(UPRO24), microalbuminuria(MAU) and creatinine in urine(Ucr) were analyzed by bio-chemical methods. The left kidneys were weighed and the right kidneys were subjected to real-time PCR analysis for the expression of CTGF and RAGE mRNA. Western blotting analysis was used to examine the expression of CTGF, RAGE, p-ERK1/2, and total-ERK1/2 protein expression. ResultsThe creatinine clearance rate, urea nitrogen, 24 h albumin excretion rate, and microalbuminuria in group D and E were significantly lower than those of group B. And the renal functions of rats in group D and E were improved and the expression of RAGE and CTGF mRNA and protein and p-ERK1/2 protein expression was significantly decreased (P<0.05). ConclusionTongluo recipe may exert its renoprotective effect through inhibiting RAGE-ERK1/2-CTGF signal pathway.

Abstract:ObjectiveTo investigate the protective effect of compound Xueshuantong capsule against damage of diabetic nephropathy in rat diabetic model. MethodsMale SD rats were given intraperitoneal injection of streptozotocin (STZ) at 60 mg/kg, and diabetic rat model was deemed successfully induced when the blood glucose level ≥16.7 mmol/L 3 days after injection. Thirty model rats were randomly divided into the following 3 groups equally: diabetic model group (DM), compound Xueshuantong capsule group (XST), and α-lipoic acid treatment group (ALA). Another 10 normal rats served as normal control group (NG). The body weight, blood glucose, blood fat, kidney weight/body weight(KW/BW), 24 h-urinary protein excretion, urea nitrogen, and serum creatinine were observed and compared between different groups after 12 weeks’ treatment. The ultrastructure changes of kidney were observed by electron microscopy and light microscope. ResultsCompared with NG group, rats in DM group had a higher 24 h-urinary protein excretion (P<0.05), and those in XST group and ALA group had a significantly lower one compared with the DM group (P<0.05). Compared with NG group, rats in the other 3 groups had a higher KW/BW, with significant difference found between DM group and XST group(P<0.05). Electron microscopic results showed uneven glomerular basement membrane thickening, foot process integration, and mesangial cell proliferation in the model group; all the above changes were improved in the XST and ALA groups. ConclusionCompound Xueshuantong capsule can lower the 24 h-urinary protein secretion, decrease KW/BW, and improve the structure of glomerular basement membrane, thus exerting renal protective effect in diabetic rats.

Abstract:ObjectiveTo compare the clinical efficacies and complications between the posterior lumbar interbody fusion(PLIF) using bilateral pedicle screw fixation and modified PLIF using unilateral pedicle screw fixation. MethodsA total of 96 patients who underwent lumbar spinal fusion operation during Feb. 2007 to May 2009 were divided into 2 groups. Group A included 40 patients (14 males and 26 female, average age being \[52.23±9.75\] years) receiving unilateral modified PLIF; Group B included 56 patients (22 males and 34 female, average age being \[56.02±10.25\] years) receiving bilateral PLIF. Oswestry disability index(ODI), visual analog scale (VAS), operating time, blood loss, length of hospital stay, hospitalization cost, fusion rates, complication rates, intervertebral height and medical expenses were all compared between the two groups. ResultsThe short-term ODI, VAS indices showed no significant difference between the two groups. Blood loss, operating time, and hospitalization cost of group A were significantly less than those in group B (P<0.05). Fusion rate was 100% in both groups 12 months after the operation. ConclusionModified PLIF using unilateral pedicle screw fixation is effective for lumbar spinal fusion, and it has the less trauma, risk and hospitalization cost compared with PLIF using bilateral pedicle screw fixation; but its long-term efficacy still needs to be further observed.

Abstract:ObjectiveTo prepare visualized iodized oil-5-fluorouracil loaded polylactic acid(PLA) micropheres for hepatic artery embolism treatment.MethodsBiocompatible and biodegradable material PLA was used as vector and iodized oil was used as positive contrast agent to prepare 5-fluorouracil loaded microspheres using double emulsion method. The preparation technology of the microspheres was developed through optimization of appearance, size distribution, drug loading, and encapsulation efficiency by orthogonal-designing method.ResultsThe prepared PLA microspheres were round in shape and had a homogenous diameter distribution. Scanning electron microscope (SEM) showed a pored surface, with an average diameter of 100 μm. The encapsulation efficiency and drug content of microspheres were (63.34%±0.54%) and (10.78%±0.14%), respectively.ConclusionWe have successfully prepared the visualized iodized oil-5-fluorouracil PLA microspheres, which can release 5-fluorouracil in a controlled manner.

Abstract:ObjectiveTo identify the effects of humid heat environment conditioning on the serum K+, Na+ concentrations and blood leukocyte count after weight-loading physical ability test (PAT), and to investigate the parameter changes with humid heat environment conditioning. MethodsA total of 59 male healthy volunteers, aged 18-23 years old, were divided into two groups, with 30 in the training group and 29 in the control group. The fasting venous blood samples were taken in the morning in both groups for determination of serum K+, Na+ concentrations and blood leukocyte count. The subjects in the training group received physical training (weight-loading) under humid heat environment condition for 6 weeks, and those in the control group received no training. Then the serum K+, Na+ concentrations and blood leukocyte count were determined again the next morning after they had participated a weight-loading PAT. Results The serum K+, Na+ concentrations and blood leukocyte counts were similar in the 2 groups before training. The increase of electrolyte concentration after PAT was more prominent in the control group compared with the baseline level before training in the training group (F=9.27, P=0.003 5; F=366.46, P<0.000 1 for K+ and Na+, respectively). Meanwhile, the control group also exhibited a significant elevation of total leukocyte count after PAT compared with the baseline level before training in the training group (F=10.20, P=0.002 3), mainly attributable to the increase of neutrophils (F=12.25, P=0.000 9). Conclusion The post-PAT serum Na+, K+ concentrations and white blood cell count, mainly neutrophils count, increase much more remarkably in the subjects undergoing no training in humid heat environment than those have been trained in humid heat environment.

Abstract:ObjectiveTo screen for differentially expressed genes associated with the development and progression of human growth hormone adenoma, so as to lay a foundation for future study. MethodsThe whole genome oligonucleotide microarray (Affymetrix 133 plus 2.0) was used to examine gene profiles of 8 growth homone adenoma samples and 2 normal pooled pituitary samples. Differentially expressed genes were subjected to bioinformatics analysis. Real-time qPCR was used to verify the microarray result of a randomly selected candidate gene. ResultsA total of 187 up-regulated genes and 899 down-regulated genes associated with growth hormone adenoma were screened out, with their functions mainly associated with molecular binding, apoptosis/tumor, metabolism, signal transduction, cell cycle, and transportation activities.Conclusion Microarray technology can be used for preliminary screen of growth hormone adenoma associated genes. The development and progression of growth homone adenoma are complex processes involving multiple genes, molecules, and pathways.

Abstract:ObjectiveTo study the in vitro antifungal activity of triazole alcohols by introduction of 1, 2, 3-triazole as the side chain.MethodsTwenty-one novel triazole alcohol compounds were designed, synthesized, and characterized by 1HNMR and MS spectra.The in vitro antifungal activities of the compounds were evaluated using eight kinds of pathogenic fungi.ResultsAll the synthesized compounds exhibited certain antifungal activities. The MIC80 value of compound 7 1-(1H-1,2,4-triazole-1-yl)-2-(2,4-difluorophenyl)-3-\[N,N-(1-substituted-benzyl-4-methylene-1H-1,2,3-triazole)\] against Candida albicans was 0.25 μg/ml, with its activity being 4 times that of fluconazole. The MIC80 value of compound Ⅵ 1-(1H-1,2,4-triazole-1-yl)-2-(2,4-difluorophenyl)-3-(N,N-dipropargyl)-2-propanols against Candida albicans was 0.015 6 μg/ml, with its activity being 64 times that of fluconazole and 4 times that of itraconazole.ConclusionThe 1,2,3-triazole can be efficiently introduced by intermolecular 1,3-dipolar cycloaddition.Large side chains may be a disadvantage for improving the antifungal activity of the title compounds.

Abstract:ObjectiveTo investigate the chemical constituents of 80% ETOH extract from rhizome of Belamcanda chinensis (L.) DC. MethodsThe constituents were isolated and purified by chromatography on silica gel, Sephadex LH-20 and HPLC; the structures of the compounds were determined by MS and NMR spectral analysis. ResultsTen constituents were identified from rhizome of Belamcanda chinensis (L.) DC, including tectoridin (Ⅰ), acetovanillone (Ⅱ), 4-hydroxy-acetophenone (Ⅲ), β-sitosterol (Ⅳ), β-daucosterol(Ⅴ), 5,7,4′-trihydroxyl-3′,5′-dimethoxyflavone (Ⅵ), luteolin (Ⅶ), apigenin (Ⅷ), 5,7,4′-trihydroxyflavanones (Ⅸ), and isorhamnetin (Ⅹ).ConclusionCompounds Ⅵ, Ⅶ, Ⅷ, and Ⅸ have been isolated from rhizome of Belamcanda chinensis for the first time.

Abstract:Adipose tissue is traditionally regarded only as an energy storage and a filler between tissues/organs. However, other important functions of adipose tissue begin to be discovered with the discovery of adipokines. Adipose tissue is also an endocrine organ, which secrets different kinds of adipokines, affecting the target organs through autocrine, paracrine and the blood circulation, and playing important roles in the pathogenesis of the obesity, insulin resistance and type 2 diabetes mellitus. This paper reviews the signal transduction pathways of adiponectin, leptin and resistin, and their changes during obesity.

Abstract:MicroRNAs(miRNAs)are a group of endogenous non-coding small RNA molecules of 21-25 nt. They can regulate gene expression at the transcriptional level through interaction with inhibition of target gene translation, participating in a variety of important biological processes. Increasing studies have found that miRNA plays a very important role in the development and progression of tumor, and that several miRNAs are closely related to the development of some tumors. Not only the miRNAs in the tissue, but also those in the peripheral blood are involved in the tumorigenesis, and they may be taken as biomarkers for diagnosis of patients, casting new lights on the early diagnosis in clinical practice. Detection of tumor-specific miRNAs in the blood is of great importance for the early diagnosis of tumors.

Abstract:Alzheimer disease (AD) is the most common form of dementia in aged population; it is a neurodegenerative disorder with unknown etiology and pathogenesis. The major pathological findings of AD include senile plaques, neurofibrillay tangles, and apoptosis-related regional synaptic and neuronal degeneration. The brain senile plaques are mainly produced by β-amyloid, a peptide with neurotoxicity and vasculotoxicity. Due to the polygenic and heterogeneous characteristics of AD, Amyloid Cascaded Hypothesis is currently drawing increasingly more attention. This paper reviews the relation between Aβ and AD published recently.

Abstract:目的应用色素原位杂交法(chromogenic in situ hybridization, CISH)检测HER2蛋白弱阳性表达乳腺癌患者的HER2基因扩增状态,为乳腺癌的临床诊治提供依据。方法收集HER2蛋白弱阳性()表达乳腺癌患者石蜡切片标本187例,采用Invitrogen公司的HER2 CISHTM 检测试剂盒,根据美国临床肿瘤学会/美国病理医师学会推荐标准,分析标本HER2基因状态及其与雌激素受体(ER)、孕激素受体(PR)表达的相关性。结果乳腺癌HER2蛋白弱阳性表达标本HER2基因扩增阳性率为53.48%(100/187),基因扩增阳性者ER、PR阳性表达率分别为44.00%、46.00%;HER2基因扩增阴性率为46.52%(87/187),基因扩增阴性者ER、PR阳性表达率分别为70.73%、78.05%。HER2基因扩增结果与性激素受体(ER、PR)表达明显负相关(P<0.01)。结论HER2蛋白弱阳性表达乳腺癌患者有必要进一步明确其基因扩增状态,CISH可有效检测此类患者的HER2基因扩增状态。

Abstract:目的评价前路减压结合椎间融合器Solis植骨融合治疗脊髓型颈椎病的临床疗效及安全性。方法31例患者中20例单节段椎间隙病变者行椎间盘摘除后植入单枚Solis椎间融合器,9例双节段椎间隙病变者和2例三节段椎间隙病变者行多节段椎间盘摘除,分别植入2枚和3枚Solis椎间融合器植骨融合,所有病例未行颈椎前路钢板固定,手术前后进行JOA评分,并观察各项疗效指标。结果术后随访6~34个月,平均(25±1.4)个月,神经症状均明显改善。术前JOA评分平均(10.4±0.5)分,术后末次JOA评分平均(14.2±0.4)分。神经功能改善率为76.3%;31例患者术后线X线片示植骨在术后12~18周获骨性融合,植骨融合率100%,术后颈椎间隙高度和生理曲度维持良好,内置物Solis-cage位置良好,无松动现象。2例患者发生脑脊液漏,3例并发喉返神经损伤,术后2个月恢复。结论颈椎前路减压椎间盘切除加单纯Solis椎间融合器植骨融合可有效治疗脊髓型颈椎病,但必须加强围手术期管理,预防并发症的发生并积极处理并发症。

Abstract:目的探讨CD4+T细胞的ATP含量与肝移植术后早期(术后1个月内)感染的相关性。方法采集85例肝移植患者、60名健康志愿者(对照组)的全血标本,术前、术后1、2、4周及感染当日和经治疗后体温正常3 d各1份;用ImmuKnowTM免疫细胞功能测定试剂盒检测CD4+T细胞的ATP值。结果感染组ATP值在术前较低,术后第1周达到低谷,显著低于同期非感染组(P<0.01)。经ROC曲线分析,术后第1周的低ATP值对诊断肝移植术后感染具有较好的敏感度和特异度;当截断值为161 μg/L时,其诊断感染的敏感度为85%,特异度为91%。感染当日的ATP含量降低与感染具有良好的相关性,经抗感染治疗体温正常3 d后,其ATP值较感染当日明显升高(P<0.01)。结论肝移植术后早期CD4+T细胞的ATP值动态变化与肝移植术后感染具有良好的相关性,监测肝移植术后患者该指标对评估其免疫状态、预防和诊治感染具有一定价值。

Abstract:目的比较不同桩核材料及漏斗深度对桩核冠修复漏斗状根管残根后的应力分布差异,寻找对残留牙体组织较少残根的最佳修复方法。方法建立桩冠修复上颌中切牙漏斗状根管残根三维有限元模型,设定漏斗状根管漏斗深度为0、2、4、6 mm。成品桩为碳纤维桩,铸造桩采用镍铬合金材料,以Von Mises 应力、最大主应力为分析指标。结果桩冠系统修复漏斗状根管的残根后根部Von Mises应力高应力区分布在唇舌侧,随漏斗的深度增加,高应力区范围逐渐扩大。与碳纤维桩相比,铸造桩修复后Von Mises应力分布不均匀,各深度组的高应力分布区较同深度碳纤维桩组明显增大,应力集中明显,根尖及漏斗底处出现了应力集中。随漏斗深度增加,碳纤维桩根牙本质Von Mises应力峰值增加了3.101 MPa,增幅为10.3%,最大主应力峰值增加了2.958 MPa,增幅为11.7%;铸造桩Von Mises应力峰值增加了6.157 MPa,增幅为22.6%,最大主应力峰值增加了5.486 MPa,增幅为19.7%。结论漏斗深度及修复材料是影响根牙本质应力的重要因素。桩核冠修复漏斗状根管的残根设计时应尽量保存根唇舌向牙体组织,以增加牙根抗力,同时在桩核冠修复设计时应降低所受咬合力。在修复漏斗状根管的残根时,尤其是深漏斗状根管的残根时,推荐使用碳纤维桩。

Abstract:目的探讨人脑胶质瘤组织CD105 mRNA和蛋白的表达及意义。方法应用组织芯片结合免疫组化技术检测152例脑胶质瘤(WHO Ⅰ级4例,Ⅱ级32例,Ⅲ级68例,Ⅳ级48例)与20例正常脑组织中CD105蛋白的表达;应用实时荧光定量PCR技术研究22例胶质瘤(Ⅱ级7例,Ⅲ~Ⅳ级15例)与4例正常脑组织中的CD105 mRNA表达。所得结果采用秩和检验、t′检验。结果脑胶质瘤组CD105 mRNA表达高于正常组(P=0.032 2),Ⅲ~Ⅳ级组高于Ⅱ级组(P<0.05)。CD105标记的微血管密度(MVD)在Ⅲ~Ⅳ级组高于Ⅰ~Ⅱ级组(P<0.05),Ⅰ~Ⅱ级组高于正常组(P<0.05)。结论人脑胶质瘤组织CD105 mRNA在转录水平已上调,CD105的表达与胶质瘤病理分级有关。
