Abstract:\[Abstract\]ObjectiveTo study the effect of rat neural stem cells on C6 glioma cell proliferation and to explore the possible mechanism.MethodsWe co-cultured neural stem cells with C6 glioma cells at different ratios (11\[2×105 2×105\], 15\[4×1042×105\],51 \[2×1054×104\]) in serum-free medium using Transwell chamber culture system for 7 days, and C6 glioma cells cultured alone served as controls. The tumorigenic ability of C6 glioma cells was observed by means of SCID mice transplantation. RT-PCR and Western blotting analysis were used to examine the expression of apoptosis-related proteins (BMP2, c-Myc, Bcl-2 and p53 mRNA) and Wnt signal molecules (β-catenin and survivin).ResultsCompared with the experimental group, the tumor cells in the tissue section of the control group had a higher malignant degree, with more mitoses, higher nuclear/cytoplasm ratio, and there were abundant single- or multi-core giant tumor cells in the tissue section. With the increase of neural stem cell proportion in the co-culture system, the tumor cell atypia degree decreased gradually, the expression of p53 mRNA in C6 glioma cells increased gradually, the expression of bone morphogenetic protein 2, Bcl-2, and c-Myc mRNA was decreased significantly (P<0.05), and the expression of β-catenin, survivin protein was decreased significantly(P<0.05). ConclusionRat neural stem cells may inhibit the tumorigenicity of C6 glioma cells by promoting apoptosis through Wnt/β-catenin pathway.

Abstract:\[Abstract\]ObjectiveTo observe the atrial expression of angiotensin Ⅱ receptor type 1(AT1R) and α7-nicotinic acetylcholine receptor(α7nAChR)during the development of heart failure in rats.MethodsMale SD rats were randomly divided into 4 groups.Abdominal aorta coarctation (AAC) was used to prepare heart failure model.Expression of AT1R and α7nAChR was determined by Western blotting analysis at 4,8,12, and 16 weeks after AAC in all the groups.ResultsCompared with the control group, expression of AT1R and α7nAChR in AAC group was not significantly different at all the 4 time points after AAC.ConclusionOur findings suggest that atrial AT1R and α7nAChR may not be involved in the early stage pathological process of AAC-induced heart failure.

    • Expression of angiotensin Ⅱ and its receptors in a rat model of chronic cyclosporine nephrotoxicity

      2010, 31(12):1286-1290. DOI: 10.3724/SP.J.1008.2010.01286

      Abstract (2889) HTML (0) PDF 2.58 M (1831) Comment (0) Favorites

Abstract:\[Abstract\]ObjectiveTo observe the expression of angiotensinⅡ(AngⅡ) and its receptors in a rat model of chronic cyclosporine (CsA) nephrotoxicity. MethodsChronic CsA nephrotoxicity was induced in Sprague-Dawley rats by administering CsA (15 mg/kg s.c.) for 4 weeks. The body weight, systolic blood pressure, serum CsA, serum creatinine (Cr), and creatinine clearance rate (Ccr) of rats were examined in each group. Trichrome staining was used to observe the tubulointerstitial fibrosis; expressions of AngⅡ and its receptors (AT1 and AT2) were examined by immunohistochemical staining and Western blotting analysis. ResultsCompared with the control rats, CsA-treated rats showed decreased body weight, increased Cr, decreased Ccr and tubulointerstitial fibrosis (P<0.01). Immunohistochemistry revealed that the immunoreactivity of AngⅡ was significantly increased in the CsA-treated rats (47±7 vs 13±4, P<0.01), with the immunoreactivity mainly locating to the juxtaglomerular afferent arterioles, and the immunoreactivity was significantly correlated with tubulointerstitial fibrosis (r=0.769, P<0.001). Western blotting analysis showed significantly decreased AT1 expression (\[114±14\]% vs \[42±6\]%, P<0.01) and increased AT2 expression (\[129±23\]% vs \[469±43\]%, P<0.01). ConclusionThe findings of our study suggest that the intrarenal renin-angiotensin system is activated during chronic CsA nephrotoxicity, which is manifested by increased AngⅡ immunoreactivity, and this increased immunoreactivity is closely related to tubulointerstitial fibrosis.

Abstract:\[Abstract\]ObjectiveTo examine the serum level of IL-22 in asthmatic patients and the expression of IL-22R1 in human airway epithelial cells, human airway smooth muscle cells, and lung fibroblasts, so as to explore the target cells of IL-22. MethodsThe serum levels of IL-22 and IL-17 in 36 asthmatic patients and 20 normal control subjects were measured by enzyme-linked immunosorbent assay (ELISA). And lung function of the asthmatic patients was assessed by Gaeger spirometry. According to the values of FEV1/FVC and FEV1%, the patients were divided into two groups: bronchodilation test positive group (19 patients) and bronchial provocation test positive group (17 patients). The expression of IL-22R1 mRNA in human airway epithelial cells, human airway smooth muscle cells, and lung fibroblasts was examined using real-time PCR, and IL-22R1 protein expression was detected by immunofluorescence staining and Western blotting analysis. ResultsThere was no significant difference in serum IL-22 and IL-17 levels between the asthmatic patients and normal controls. The serum IL-22 and IL-17 levels in asthmatic patients positive for bronchodilation test was significantly higher than those positive for bronchial provocation test(P<0.05). IL-22R1 mRNA and protein were detected in all the 3 types of cells. ConclusionIL-22 may be involved in the pathogenesis of asthma, and human airway epithelial cells, human airway smooth muscle cells, and lung fibroblasts may all be the target cells of IL-22.

Abstract:\[Abstract\]ObjectiveTo investigate the effect of insulin-like growth factor-1 (IGF-1) on expression of cystationine-β-synthase(CBS)and hydrogen sulfide(H2S) in rat adrenal pheochromocytoma cell line (PC12) and the possible signaling pathway. MethodsAfter PC12 cells were treated with 20,40, or 80 ng/ml IGF-1 for 24 h,fluorecence quantitative PCR was used to examined the expression of CBS gene, Western blotting analysis was used to detect the expression of CBS,ERK/MAPK protein expression, and sensitive sulphur electrode was used to analyze the levels of H2S gas. For inhibition studies,25 or 50 mol/L PD98059,a specific ERK/MAPK inhibitor, was used 30 min prior to 80 ng/ml IGF-1 treatment; and the above parameters were examined again using the same method. ResultsIGF-1 increased the expression of CBS,pERK1/2, and the levels of H2S;and PD98059 could inhibit the above effect of IGF-1.ConclusionIGF-1 and its ERK/MAPK signaling pathway regulate the expression of CBS and levels of H2S.

Abstract:\[Abstract\]ObjectiveTo investigate the relation of diabetic encephalopathy with oxidative stress and hippocampus expression of inducible nitric oxide synthase(iNOS) in rats and to study the therapeutic effects of curcumin. MethodsDiabetes mellitus was induced by injection of Streptozotocin in rats. The experiment animals were randomly divided into control group, diabetic group, diabetes plus curcumin group and control plus curcumin group; animals in the latter two groups received 60 mg·kg-1·d-1 curcumin treatment. Superoxide dismutase(SOD),catalase(CAT), malondialdehyde(MDA) and nitric oxide(NO) in the hippocampal tissues were measured 12 weeks after treatment. The expression of iNOS mRNA and the mean optical density (MOD) of immunoreactive neurons were evaluated by RT-PCR and immunohistochemistry method, respectively. ResultsCompared with the control group, diabetic group had significantly lower activities of SOD and CAT, higher contents of NO and MDA, and increased expression of iNOS mRNA and protein in the hippocampal tissues (P<0.01). Compared with the diabetic group, diabetic plus curcumin group had significantly higher activities of SOD and CAT, lower contents of NO and MDA and decreased expression of iNOS mRNA and the MOD of immunoreactive neurons in hippocampus (P<0.01). ConclusionDiabetic encephalopathy is associated with oxidative stress and increased expression of iNOS; curcumin may protect diabetic encephalopathy by inhibiting oxidative stress and down-regulating the iNOS expression.

Abstract:\[Abstract\]ObjectiveTo study the effect of puerarin (Pue) on hypertrophy and apoptosis of neonatal rat cardiomyocytes induced by isoproterenol(ISO). MethodsCardiomyocytes of neonatal rats were cultured in vitro and were treated with ISO(10-5mol/L), Pue(10-6mol/L), ISO(10-5mol/L)+ Pue(10-6mol/L), or ISO(10-5mol/L)+propranolol (Pro, 2×10-6 mol/L); untreated cells were taken as control. The total protein content, cardiomyocyte volume, protein synthesis and apoptosis rate of the cardiomyocytes were investigated. ResultsCompared with the control group, ISO significantly increased the total protein content, cardiomyocyte volume and apoptosis rate of cardiomyocytes (P<0.01).Similar to Pro, Pue inhibited the above changes induced by ISO. ConclusionPue can inhibit the hypertrophy and apoptosis of cardiomyocytes induced by ISO, which might be one of the reasons for the protective effect of Pue against myocardial hypertrophy.

    • Correlation of survivin gene with hepatocellular carcinoma in Han nationality in east China

      2010, 31(12):1314-1318. DOI: 10.3724/SP.J.1008.2010.01314

      Abstract (2120) HTML (0) PDF 375.69 K (2075) Comment (0) Favorites

Abstract:\[Abstract\]ObjectiveTo explore the association of survivin gene with hepatocellular carcinoma(HCC) in Han nationality in east China. MethodsA case-control study was designed, which included 176 HCC cases and 196 healthy controls, all from Haimen city of Jiangsu province. The rs1042489 and rs9904341 loci of survivin gene were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique, and the association of the genotypes and haplotypes of the two loci with HCC was analyzed under different genetic models. ResultsUnivariate analysis showed no significant difference in the genotypes or allele distribution between HCC case group and the control group under different genetic models in either rs9904341 or rs1042489 loci (P>0.05). Linkage disequilibrium (LD) analysis showed that the two loci were in LD (χ2=4.777,P=0.03), with the D’ value being 0.188 and 0.183 in the case and control group, respectively. The recessive genetic model was found to be the optimal model by multivariate Logistic regression, according to the lowest value of Akaike’s information criteria (AIC). With no rs9904341C-rs1042489T (C-T) haplotype as reference, it was found that the haplotype of C-T from the two loci was associated with a lower risk for HCC under the recessive genetic model, with the factors such as alcohol drinking and HBV infection controlled (OR=0.48, P=0.04), and no other haplotypes were found associated with HCC. Multivariate analysis showed that HBsAg+ and hepatitis B were the risk factors for HCC, but without haplotype-environment interaction. ConclusionNo association has been found for rs9904341 or rs1042489 in survivin gene with HCC in Han nationality in east China, and rs9904341C-rs1042489T might be a protective haplotype for HCC.

Abstract:\[Abstract\]ObjectiveTo investigate the endothelial function of chronic ischemia heart failure (CIHF) patients complicated with depression. MethodsThe depression statuses were evaluated by Self-rating Depression Scale (SDS) in 56 patients with documented stable chronic ischemia heart failure. The patients were assigned to depression group (n=20) or non-depression group(n=36) based on their standard SDS scores. The flow-mediated dilation(FMD) of the brachial artery was assessed by ultrasound in the two groups, and the plasma ET-1 and NO levels were determined so as to evaluate the endothelial function. ResultsThe incidence of depression was 35.7% in the 56 patients. Patients in the depression group showed significantly lower FMD compared with the non-depression group(\[5.97±0.78\]% vs \[6.66±0.83\]%, P<0.05). Plasma NO level (\[48.90±9.82\] μmol/L vs \[55.13±10.32\] μmol/L, P<0.05) was lower and ET-1 level (\[83.35±13.39\] ng/L vs \[74.67±10.95\] ng/L, P<0.05) was higher in the depression group than in the non-depression group.ConclusionCIHF patients with depression have a more severe damage of endothelial function than those without depression.

      2010, 31(12):1323-1325. DOI: 10.3724/SP.J.1008.2010.01323

      Abstract (2162) HTML (0) PDF 1.46 M (2376) Comment (0) Favorites

Abstract:\[Abstract\]ObjectiveTo use ultrasound elasticity imaging (UEI) technology for evaluating the elasticity changes of the deep vein thrombosis (DVT) at different stages. MethodsConventional ultrasonic diagnoses were performed in 49 patients with DVT and the patients were divided into acute stage, sub-acute stage, and chronic stage. The elastic image characteristics of the thrombosis were observed at different stages and the strain ratio (B/A) between the muscle tissue around the vein and the thrombosis was calculated. Results(1) In ultrasound elasticity image, the acute thrombosis was mainly red, the sub-acute thrombosis was green, and the chronic thrombosis was blue. (2)The strain ratio of the sub-acute thrombosis group and the chronic thrombosis group were significantly higher than that of the acute thrombosis group(P<0.001, P<0.05), and the strain ratio of the chronic thrombosis group was significantly higher than that of the sub-acute thrombosis group(P<0.05). ConclusionUEI technology can be used to evaluate the elastic changes in different stages of the deep vein thrombosis, and it can help to select treatments for thrombosis and predicting the complications.

    • QRS dispersion in assessment of left ventricular systolic function and asynchronous left ventricular wall motion in patients with left buddle-branch block

      2010, 31(12):1326-1329. DOI: 10.3724/SP.J.1008.2010.01326

      Abstract (3332) HTML (0) PDF 268.13 K (2399) Comment (0) Favorites

Abstract:\[Abstract\]ObjectiveTo evaluate the role of QRS dispersion(QRSD) in evaluating left ventricular function and the asynchrony of ventricular motion in patients with left buddle-branch block(LBBB). MethodsForty-two hospitalized patients with LBBB and 28 normal controls (group C) were included in the present study. The 42 LBBB patients were further divided into 2 groups: the isolated LBBB patients (group A, n=7) and LBBB patients with other organic diseases (group B, n=35). The indices of echocardiography and electrocardiogram were compared and analyzed among the three groups, including left ventricular ejection(LVEF), left ventricular end-diastolid dimension(LVEDD), E/A ratio, interventricular mechanical delay(IVMD), left ventricular systolic asynchrony index (TS-SD), QRSD, and QRS duration(QRSd). Multivariate correlation analysis was used to analyze the correlation of QRSD and QRSd with the indices of electrocardiogram and echocardiography in the two LBBB groups. ResultsAll the indices were significantly different between group B and group C (P<0.05), except for LVEDD in group A (P=0.145). All the indices were significantly different between group A and group B, except for QRSd (P=0.175). The QRSd and QRSD were negatively correlated with LVEF in the two LBBB groups, with the correlation coefficients being -0.310 (P=0.046) for QRSd and -0.341 (P=0.027) for QRSD; QRSD and TS-SD were positively correlated, with the correlation coefficient being 0.318 (P=0.040). The rest indices were not significantly correlated. ConclusionQRSD of ECG can be used for evaluating left ventricular systolic dysfunction in patients with LBBB. However, further studies are needed in order to determine its role in estimating the asynchrony of ventricular motion.

Abstract:\[Abstract\]ObjectiveTo investigate the effects of dexmedetomidine on blood glucose, β-endorphin(β-EP), tumor necrosis factor-α(TNF-α) and interleukin-6 (IL-6) levels in patients undergoing radical esophagectomy, and to discuss the possible mechanism. MethodsForty patients with esophageal carcinoma undergoing radical esophagectomy were randomly assigned to 2 groups: dexmedetomidine group and control group (n=20 in each). Blood samples were taken immediately before induction of anesthesia(T0), immediately after tracheal extubation(T1), day 1 and day 2 after operation(T2,T3) for determining the levels of blood glucose, plasma β-EP and serum TNF-α and IL-6. ResultsCompared with those at T0, MAP and HR at T1were significantly elevated (P<0.05) in the control group, the blood glucose and plasma β-EP at T1-T3were significantly increased (P<0.05), and the serum TNF-α and IL-6 at T1 and T2 were significantly elevated(P<0.05). The blood glucose, plasma β-EP, serum TNF-α and IL-6 in dexmedetomidine group had no significant changes at all time points (P>0.05), and the above parameters were significantly different between the two groups at corresponding time pionts (P<0.05). ConclusionContinued application of dexmedetomidine can effectively decrease the levels of blood glucose, plasma β-EP, serum TNF-α and IL-6 in patients undergoing radical esophagectomy, inhibiting the peri-operative stress response.

Abstract:\[Abstract\]ObjectiveTo conduct a systematic review on the clinical effectiveness and relative efficiency of stapled and unstapled mesh in laparoscopic inguinal hernia repair. MethodsPubMed, EMBASE, CENTREN, Cochrane Central Register of Controlled Trials (CENTRAL), CBMdisk, and National Knowledge Infrastructure(CNKI) database were searched for randomized controlled trials comparing stapled and unstapled mesh in the laparoscopic repair of inguinal hernia; manual search was also conducted. The obtained literatures were rated and eligible trails were subjected to meta-analysis using RevMan5.0 software. ResultsEight trials assessing 1,619 participants were finally included. Meta-analysis showed that there were no significant differences between the compared arms in post-operation recurrence rate (RR=1.24,95%CI 0.36-4.26,P=0.74),total complication (RR=1.18, 95% CI 0.84-1.65, P=0.34 ), time to return to normal activity(MD=0.14, 95%CI -0.16-0.44,P=0.37). The mesh staple group needed more operative time(MD=4.52, 95%CI 1.46-7.59,P=0.004), cost more, and there was no definite conclusion on the post-operative pain in the two groups.ConclusionThere is no noticeable difference in the clinical effectiveness between the stapled and unstapled mesh in laparoscopic inguinal hernia repair; the stapled mesh costs more, needs more time; and the post-operative pain still needs to be studied by more well-designed and large sample randomized controlled trials.

Abstract:\[Abstract\]ObjectiveTo investigate the association of serum leptin level with erectile dysfunction (ED). MethodsA total of 38 out-patients complaining ED were graded by using IIEF-5 questionnaire. The mean IIEF-5 score of the patients was (12.3±2.4)and no one was higher than 21. The patients were divided into 3 groups according to their scores. The fasting serum leptin levels were determined by ELISA in 38 patients (patient group) and 15 healthy controls (control group). Their body weights and heights were measured for body mass index(BMI). ResultsSerum leptin levels were significantly correlated with BMI in both groups, regardless of patient ages and history. The mean leptin level in patients was (10.16±8.67) μg/L, which was significantly higher than that in the healthy controls (\[4.81±3.66\] μg/L,P<0.05) after adjusting the effect of BMI. The leptin levels in the mild, moderate, and severe ED groups were (6.98±4.99) μg/L, (10.3±9.51) μg/L and (13.1±10.02) μg/L, respectively, with that of the mild group being significantly lower than those of the latter two groups(P<0.05). Conclusion It is indicated that human serum leptin level may have a potential value in diagnosing ED.

Abstract:\[Abstract\]ObjectiveTo summarize our experience and operative techniques of transumbilical laparo-endoscopic single-site partial nephrectomy (TLSPN). MethodsTLSPN was performed on two patients for the right kidneys in August and September of 2009. ResultsThe operation was successfully done in both cases, with an additional trocar of 5 mm to retract the liver. The operation time was 255 min and 240 min; the time periods of renal artery occlusion were 48 min and 40 min; and the intraoperative blood losses were 100 ml and 50 ml. None of the patients received blood transfusion or antalgic. The drainage was removed 9 days and 5 days after operation, and the postoperative hospital stays were 13 and 12 days. ConclusionTLSPN is a safe and effective method with less trauma. However, special instruments are needed for the procedure, and the suture and knotting are somewhat difficult. An assistant trocar is needed for the procedure for the right kidney.

Abstract:\[Abstract\]ObjectiveTo assess the clinical effect of modified Mapleson D breathing system in intravenous anesthesia for minor surgical procedures. MethodsA total of 112 patients (ASA Ⅰ-Ⅱ) scheduled for minor outpatient surgery were randomly divided into two groups, with 56 in each group. Patients in Group A inhaled oxygen using common face mask, and were transferred to artificial ventilation by emergency ventilator when respiratory depression occurred; those in Group B were subjected to modified Mapleson D breathing system, and were transferred to artificial ventilation using the present breathing system when respiratory depression occurred. The respiratory rate (RR), oxygen saturation (SpO2), Partial end-tidal CO2 pressure (PETCO2), occurrence (T1) and rectification time (T2) of respiratory depression were all recorded in the two groups.ResultsThe incidence of respiratory depression in Group B was lower than that in Group A; moreover, Group B also had a higher incidence of hyoxemia and shorter rectification time compared with Group A. ConclusionCompared with conventional face mask, our modified Mapleson D breathing system is easy and convenient to use, and it leads to lower incidence and quick rectification of respiratory depression, showing a potential in the airway management of patients undergoing minor intravenous anesthesia.

Abstract:\[Abstract\]Recently increasing functions of nitric oxide (NO) has been discovered. Researchers around the world have reported that NO plays an important role in the anti-microbial infection; however, more recent studies have found that NO also protect the growth of microorganisms, for example, it can facilitate drug resistance of bacteria and it can protect microorganisms from oxidative stress and pressure. This paper reviews both positive and negative roles of NO on microbial growth and the related mechanisms.

Abstract:\[Abstract\]Danger associated molecular pattern(DAMPs) is defined as the danger signal molecules that can be recognized by pattern recognition receptors (PRRs).High-mobility group box 1 protein (HMGB1) belongs to DAMPs and the release of HMGB1 indicates the damage of tissues. Many researches have showed that HMGB1 participates in tumor angiogenesis, evasion of apoptosis,self-sufficiency in growth signals and invasion and metastasis, indicating HMGB1 can promote tumor development and progression.More recent findings indicate that HMGB1 can be used as an adjuvant to enhance the efficiency of the antitumor immune response.Various reports have suggested that HMGB1 plays a double phase action in tumor immunological regulation. This papers is aimed to review the role of HMGB1 in tumor immunological regulation and the related mechanisms.
