Abstract:ObjectiveTo investigate the relationship of excision repair cross-complementing 1(ERCC1),xeroderma pigmentosum group D(XPD),and breast cancer susceptibility gene 1(BRCA1) polymorphisms with the efficacy of platinum-based chemotherapy for treatment of the patients with advanced non-small cell lung cancer.MethodsA total of 124 patients with advanced NSCLC were routinely treated with platinum-based chemotherapy,and their clinical responses were evaluated.ERCC1 Asn118Asn(rs11615),XPD Lys751Gln(rs13181) and BRCA1 Ser1613Gly(rs1799966) of the patients were genotyped using the TaqMan method.The association of ERCC1 Asn118Asn,XPD Lys751Gln and BRCA1 Ser1613Gly polymorphisms with the patient responses was analyzed using unconditional logistic regression model.ResultsIt was found that the BRCA1 Ser1613Gly polymorphism was significantly correlated with clinical benefit(P=0.014).Patients carrying Gly allele had better clinical benefit than patients with wildtype allele(P=0.006).No significant association was found between ERCC1 and XPD polymorphisms with clinical benefit.Furthermore,we found that the three SNPs in NER(nucleotide excision repair) could work together.More variant alleles(ERCC1 T,XPD Gln and BRCA1 Gly) was associated with better clinical benefit(P=0.036).ConclusionThe BRCA1 Ser1613Gly polymorphism of NER is associated with the clinical benefit of NSCLC patients receiving platinum-based chemotherapy.Analysis of SNPs of more genes may help to guide drug choosing for chemotherapy.

Abstract:ObjectiveTo investigate the differentially expressed genes and their regulatory network during priming phase of liver regeneration in mice using gene chip technology,bioinformatic analysis,and chromatin immunoprecipitation methods.MethodsLiver tissues of mice from post-hepatectomy and sham groups were processed for microarray to detect the differentially expressed genes.PAINT analysis based on TRANSFAC database was used to analyze the transcription regulatory network for differentially expressed genes.Chromatin immunoprecipitation was employed for the verification of the target genes of NF-κB.ResultsA total of 332 genes were differentially expressed during the priming phase of liver regeneration,with 205 down-regulated and 127 up-regulated.The immediate early response 2(IER2) was increased by about 5 folds.PAINT analysis showed that several transcription factors such as Stat family and NF-κB participated in the regulatory network of these genes.Chromatin immunoprecipitation demonstrated that IER2 was regulated by NF-κB.ConclusionNF-κB participates in priming phase of liver regeneration through regulating IER2.

Abstract:ObjectiveTo investigate the physicochemical properties and biological activity of self-prepared fusion protein inducible co-stimulator-Ig. MethodsAcid hydrolysis, edman degradation and peptide mass finger printing were used to determine the amino acid composition, N-terminal 15 amino acid sequences, and peptide mapping. In vivo mixed lymphocyte reaction assay was used for identification of its biological activity. ResultsThe result of amino acids composition analysis was consistent with the theoretical value of ICOS-Ig. N-terminal 15 amino acid sequences of the product were EINGSANYEMFIFHN, consistent with the theoretical value of ICOS-Ig. Peptide match assay identified six peptides of the product which could match the theoretic maps of ICOS-Ig. ICOS-Ig and CsA noticeably inhibited the proliferation of allo-reactive T cells in vivo. ConclusionThe prepared ICOS-Ig fusion protein has a correct structure and can inhibit the proliferation of allogeneic T cells in vivo, which lays a foundation for quality control of ICOS-Ig fusion protein.

Abstract:ObjectiveTo investigate the influence of TTS-12, a steroid saponin from Tribulus terrestris L., on the formation of Candida albicans biofilm, and to discuss the possible mechanism. MethodsThe inhibition of C.albicans biofilm formation by TTS -12 was observed by confocal scanning laser microscopy; XTT method was used to investigate the influence of different concentrations of TTS-12 on C.albicans biofilm formation. The water-hydrocarbon two-phase assay was used to measure the cell surface hydrophobicity of C.albicans treated with different concentrations of TTS-12. The expression of CSH1 mRNA was measured by real-time RT-PCR. ResultsCompared with control group, TTS-12 treatment resulted in loose C.albicans biofilm,and it dose-dependently inhibited C.albicans biofilm formation. The cell surface hydrophobicity in the TTS-12-treated groups was lower than that in the control group; consistent with this, TTS-12-treated cells also expressed significantly lower levels of CSH1 mRNA than cells in control group (P<0.01). ConclusionTTS-12 may inhibit the formation of C.albicans biofilm through inhibiting CSH1 gene expression.

Abstract:ObjectiveTo evaluate the effect of pretreating donor liver with salvianolate on biliary injury after orthotopic liver transplantation (OLT) in rats,and to discuss the underlying mechanisms. MethodsAccording to the preservation solutions of the liver grafts, the recipients rats were allocated in three groups: UW solution group, salvianolate group (UW solution + 0.4 mg/ml salvianolate), and saline group (UW solution + saline). In experiment 1, the recipient rats underwent arterialized OLT with biliary extra-drainage, and the bile flow and bile salt secretion were recorded at 1, 6, and 24 h post-transplantation. In experiment 2, the recipient rats underwent arterialized OLT, and the serum alkaline phosphatase, bile salt, total bilirubin, and the incidence of biliary complications were examined at 2 weeks post-transplantation. Meanwhile, the mRNA and protein expression of bile salt export pump (BSEP) in the liver samples were also detected using real-time RT-PCR and Western blotting analysis,respectively. ResultsCompared with the UW solution and saline groups, salvianolate group exhibited significantly increased bile flow and bile salt secretion at 1, 6, and 24 h post-transplantation (P<0.05). At 2 weeks post-transplantation, salvianolate group had significantly lower serum alkaline phosphatase, bile salt, total bilirubin(P<0.05), and incidence of biliary complications(P<0.01), and significantly higher expression of BSEP mRNA and protein compared with the other two groups (P<0.05). ConclusionSalvianolate addition in UW preservation solution can ameliorate biliary injury and reduce the incidence of biliary complications after OLT in rats, which might be associated with increased expression of BSEP in liver graft.

Abstract:ObjectiveTo explore the role of c-Jun NH2 terminal kinase (JNK) activation and JNK-mediated apoptotic signal pathway in intestinal ischemia/reperfusion (II/R) in mice.Methods C57BL/6 mice were randomly divided into sham-operated group (n=6) and II/R groups (n=36); the latter was further divided according to time after perfusion (0,0.5,1,4,6 and 12 h).Animal II/R model was established by clamping the superior mesenteric artery (SMA) for 40 min followed by reperfusion.Animals in the sham-operated group received no clamping.Animals in the two groups were sacrificed at defined time points,and the expression of JNK,phosphorylation (phospho-) JNK,cleaved caspase-3,Bcl-2 and Bax protein in the intestinal tissue was examined by Western blotting analysis,and the pathological changes of ileum tissue were observed under optical microscope.Results Most severe intestinal injury was found at the early stage of reperfusion,and the intestinal tissues almost recovered 12 h later.The phospho-JNK in the intestine was significantly elevated within 1 h after II/R compared with sham group (P<0.01).Cleaved caspase-3 was significantly increased in II/R group at 0.5 h,1 h after reperfusion compared to sham group (P<0.01); the expression of Bcl-2 protein in II/R group was significantly decreased compared with the sham-operated group (P<0.01),and there was no significant difference in Bax expression between different groups.Conclusion JNK phosphorylation plays an essential role in the intestinal damages induced by II/R,possibly through down-regulating Bcl-2 protein expression and caspase-3 dependent apoptosis pathway.

Abstract:ObjectiveTo investigate the protective effect of deferoxamine preconditioning against hypoxia injury in astrocytes and the underlying mechanisms. MethodsAstrocytes were cultured under ischemia stress, which was mimicked by oxygen and glucose deprivation (OGD) with deferoxamine. Astrocytes were divided into three groups: normally cultured group; Deferoxamine pretreated group: astrocytes were pretreated with Deferoxamine and then treated with Deferoxamine OGD; and OGD group; astrocytes were treated with Deferoxamine OGD. The cell viability examination, the ratio of condensed nuclei, and the morphological changes were used to assess the protective effect of deferoxamine. The expression of HIF-1α and EPO protein and mRNA in astrocytes was examined by immunofluorescence staining and RT-PCR, respectively. ResultsThe morphology of AS in the deferoxamine pretreated group kept intact. AS viability in deferoxamine pretreated group was 58% of the normally cultured group, and that in the OGD group was 25% of the normally cultured group (P<0.05). The ratio of condensed nuclei in deferoxamine pretreated group was 38% and that in OGD group was 30%(P<0.05).Immunofluorescence staining found that expression HIF-1α, and EPO protein appeared in cultured astrocytes after deferoxamine pretreatment. RT-PCR confirmed that mRNA of HIF-1α and EPO was up-regulated after deferoxamine pretreatment. ConclusionDeferoxamine preconditioning can protect the astrocytes from hypoxia damage, which is related to deferoxamine-induced increase of HIF-1α and EPO expression.

Abstract:ObjectiveTo investigate the changes of XAB2 mRNA and protein expression in gastric cancer tissues, so as to discuss its possible relationship with the development of gastric cancer. MethodsXAB2 mRNA expression in 34 gastric cancer tissues and the matched adjacent normal tissues was examined by real-time RT-PCR. Meanwhile, tissue biochip and immunohistochemistry were employed to examine XAB2 protein expression in gastric cancer tissues, matched adjacent normal tissues, and gastritis tissues. The expression of XAB2 in different tissues and its clinical significance was analyzed. Results XAB2 mRNA expression in the gastric cancer tissues was significantly lower than that in corresponding adjacent normal tissues (P<0.01), and the expression level was greatly decreased at early stage of gastric cancer (stageⅠ+Ⅱ). Immunohistochemical examination showed that the positive rate of XAB2 gene was significantly lower in gastric cancer tissues (50.0%, 17/34) than those in the adjacent tissues (82.4%, 28/34) and gastritis tissues (91.1%, 31/34, P<0.01),with no significant differences found between the latter two tissues. The XAB2 protein expression in gastric cancer tissues was positively correlated to tumor differentiation(P=0.041) and negatively with tumor infiltration and size (P=0.019 and P=0.027, respectively). XAB2 expression was not associated with patients’ sex, age, tumor position, lymph node metastasis or TNM stages(P>0.05). Conclusion The lower XAB2 expression might be closely associated with the development and progression of gastric cancer, and further studying may benefit the diagnosis and therapy of gastric carcinoma patients.

Abstract:ObjectiveTo establish a single-tube nested multiplex-PCR assay for rapid detection and typing of Dengue viruses for multiple infections with different serotypes. MethodsA pair of outer universal primers designed for all the four Dengue virus serotypes were used to amplify the mixed RNA of 1-4 dengue viral serotypes by one-step RT-PCR,and the products were used as template for nested multiplex PCR using four pairs of serotype-specific primers in the same reaction tube. The sensitivity and specificity of single-tube nested multiplex PCR assay amplifying from the mixed 1-4 serotype dengue viral RNA were subsequently compared with those amplifying from the single serotypes dengue viral RNA. ResultsBy optimizing the reaction condition,four specific fragments (482,119,290,and 389 bp) were successfully amplified from the mixed RNA of 1-4 serotypes dengue viruses in single tube by single-tube nested multiple-PCR. Its sensitivity and specificity amplifying from the mixed RNA of 1-4 serotypes dengue viruses were similar to those amplifying from the single serotype dengue viral RNA. The detection limit of nested multiple-PCR was 66.068 copies/μl. ConclusionSingle-tube nested multiple-PCR method is simple,rapid,sensitive, and specific for detecting and typing dengue viruses,and it is valuable for detecting and typing of the clinical multiple infections.

Abstract:ObjectiveTo study the effect of metastasis suppressor gene Kai1/CD82 on the cell proliferation of Hep-2 cell line.MethodsRecombinant adenovirus rAd-Kai1 was amplified,purified,and was used to transfect laryngo-carcinoma Hep-2 cell line.Cells transfected with blank vector and untransfected cells served as controls.MTT assay was used to assess the influence of Kai1 gene on proliferation of Hep-2 cells; RT-PCR was used to examine the expression of Kai1 gene in cells of different groups.ResultsThe titer of rAd-Kai1 reached 6×1010PFU/ml after expansion and purification.When the concentration of the adenovirus was 30 MOI,the transfection rate of Hep-2 cells could be higher than 90%.MTT assay showed that the cell proliferation abilities were similar in the two control groups,and the proliferation ability of cells in the Kai1 transfection group was significantly lower than that in the non-transfected group (P<0.05),with the inhibitory rate in the Kai1 group being 29%.ConclusionThe Kai1/CD82 gene can inhibit the proliferation of Hep-2 cells.

Abstract:ObjectiveTo observe the types, prevalence of cardiac dyssynchrony in patients of chronic systolic heart failure (SHF) , and to investigate the value of QRS duration >135 ms in selecting patients for cardiac resynchronization therapy (CRT). MethodsForty-five patients with SHF were randomly divided into two groups according to QRS width: Group 1 (QRS width <120 ms, 21 patients) and Group 2 (QRS width ≥120 ms, 24 patients). Electrocardiography, conventional echocardiography, and tissue doppler were performed in all patients to evaluate the cardiac systolic dyssynchrony. Meanwhile, the diagnostic value of optimized QRS width to evaluate the dyssynchrony indices was tested while using 135 ms as new cutoff value. ResultsSystolic function: compared with Group 2, Group 1 had significantly higher LVEDD and LVEDV(P=0.03 and 0.01, respectively), lower LVEF(P=0.004), and had significantly higher NYHA class(P=0.002)and more frequent use of diuretics and digoxins(P=0.01,both). Dyssynchrony parameters: Group 2 had a significantly higher prevalence of interventricular dyssynchrony(70.8% vs 15%,P<0.000 1) and higher IVMD value(P<0.000 1) than Group 1; the prevalences of intraventricular dyssynchrony were high in both groups(71.4% vs 79.2%,P=0.55), except that the APET value was prolonged in Group 2. Other indices(Ts-SD, Ts-12, and SPWMD)showed no significant difference between the two groups. Correlation analysis: a strong correlation was found between QRS duration and interventricular dyssynchrony(r=0.58,P<0.000 1), while it was fairly poor between QRS duration with intraventricular dyssynchrony(r=0.07,P=0.64).ROC analysis: QRS width >135 ms could finely predict the presence of interventricular dyssynchrony, with a sensitivity of 80% and a specificity of 87.5%; while the same cutoff value to predict intraventricular dyssynchrony only yielded a sensitivity of 44.1% and specificity of 73.6%.ConclusionIntraventricular dyssynchrony and(or) interventricular dyssynchrony has a high prevalence in patients with SHF. A QRS duration cutoff value higher 135 ms can well evaluate the cardiac mechanical dyssynchrony in clinical practice, which may be of value for optimizing selection of CRT candidates and reducing the nonresponders.

Abstract:ObjectiveTo identify the optimal rotation time and pitch of 64-slice spiral CT angiography for arterial occlusive diseases (AOD) of the lower limbs. MethodsWe performed test-bolus in eighty patients with AOD of the lower limbs (48 with intermittent claudication and 32 with ischemic rest pain or ulceration) to confirm the aortic peak time, popliteal artery peak time and aortopopliteal bolus transit time. The patients were randomly assigned to receive two sets of scan protocols for lower limb arterial 64-slice spiral CT angiography (CTA) examination (n=40): protocol A, with a gantry rotation time of 330 ms and a pitch of 1.0; protocol B, with a gantry rotation time of 500 ms and a pitch of 0.85. Two experienced radiologists independently evaluated the image quality of the lower limb artery. ResultsGreat differences in the time to peak enhancement in the aorta (14-33 s) and popliteal arteries (20-48 s) and the aortopopliteal bolus transit time (4-24 s) were found between different AOD patients. Wide overlap of the time to peak enhancement and transit time was observed between intermittent claudication patients and ischemic rest pain or ulceration patients. The qualities of segment images were rated as excellent 63.3% (152/240), good 30.0% (72/240), and poor 30.0% (72/240) for protocol A, and excellent 98.3% (234/238) and good 1.7% (4/238) for protocol B, with no poor images in protocol B. Conclusion64-slice CTA with a gantry rotation time of 500 ms, a pitch of 0.85, scan time>30 s, and with the contrast agent injection time maintained at 35 s, can obtain better image of lower limb arteries in patients with AOD of the lower limbs.

Abstract:ObjectiveTo analyze the correlation of intervetebral disc degeneration with the diffusion values of lumbar vertebral discs and lumbar blood supply in patients with suspected lumbar intervertebral problems, so as to assess ADC value in diagnosis of intervertebral degeneration and the related blood supply.MethodsSixty-five patients with suspected lumbar intervertebral problems were included in the present study; they had a mean age of (52.3±28.5) years and an age range of 16-76 years. Eleven of them had low back pain complicated with sciatica, 16 had low back pain without sciatica, 33 had sciatica without low back pain, and 5 had intermittent claudication. The disease course was 1-57 weeks, with a mean of (13.4±6.3) weeks. MR angiography and diffusion imaging were performed to evaluate the status of lumbar arteries and to obtain the ADC values of L1-4, so as to analyze the correlation of ADC values, lumbar blood supply, and intervetebral disc degeneration.ResultsThe ADC values of L1-4 were negatively correlated with the degeneration of lumbar discs(P<0.05). There was a relation between the artery status of L1, L2, and L3 with the degeneration of corresponding lumbar discs of L1-2, L2-3, and L3-4(rs=0.823,P=0.016 3; rs=0.791, P=0.019 2; rs=0.835, P=0.010 3 ). The artery status of L4 and the L4-5 discs was not significantly correlated with degeneration of corresponding lumbar discs(rs=0.306, P=0.209 2).ConclusionThe ADC values of lumbar intervertebral disc can reflect the degeneration of L1-4, and degeneration of L1-2, L2-3, and L3-4 is correlated with the artery status of corresponding discs.

Abstract:ObjectiveTo study the protective effect of coenzyme complex on myocardium under cardiopulmonary bypass in open heart surgery.MethodsForty patients with congenital heart disease scheduled for open heart surgery under cardiopulmonary bypass (CPB) were evenly randomized into 2 groups. Coenzyme complex (1 bottle/10 kg) was given to the experimental group but not to the control group. The venous blood samples were collected at following time points. T1: during anesthetic induction period (baseline), T2: half an hour after aorta clamp, T3: one hour after aorta clamp opened, T4: 24 h after operation, T5: 48 h after operation, T6:72 h after operation, and T7:one week after operation. The levels of cardiac troponin I (CTnI), interleukin-8(IL-8) and maleic dialdehyde (MDA) were measured. Right atrial appendage specimen was taken and processed for transmission electron microscope observation of myocardial ultrastructure.ResultsThe levels of CTnI, IL-8, and MDA were significantly increased in the two groups during CPB, but the values in the experimental group were lower than those in the control group at following time points: (CTnI) one hour after aorta clamp opened and 24 h, 48 h, 72 h after operation (P<0.05 or 0.01); (IL-8) half an hour after aorta clamp, one hour after aorta clamp opened and 24 h, 48 h, 72 h after operation (P<0.01); and (MDA) one hour after aorta clamp opened and 24 h, 48 h after operation (P<0.05 or 0.01). The injury of myocardial cellular ultrastructure was seen in each group after aorta clamp was opened by different extents. However, more severe disorganization of myocardial cellular ultrastructure could be seen in the control group. ConclusionCoenzyme complex may reduce the production of oxygen free radical and inflammatory factor, subsequently reduce the release of CTnI, protecting myocardium under cardiopulmonary bypass in the open heart surgery.

Abstract:ObjectiveTo construct a 5/11 chimeric adenovirus harboring RGD-4C,and observe its transfection efficiency after transfecting it into hepatocarcinoma cell lines HepG2,BEL-7404 and BJ cells.MethodsRGD-4C was inserted into the HI loop region of 5/11 chimeric adenovirus fiber gene and the insertion outcome was confirmed by enzyme digestion and PCR analysis.The confirmed plasmid was co-transfected into E.coli BJ5183 together with adenoviral backbone plasmid pPE3 to produce recombinant plasmid by homologous recombination.Recombinants were selected and co-transfected into 293 cell line with PDC328-EF1-EGFP to produce recombinant adenovirus.The recombinant adenovirus production was confirmed by PCR analysis and was amplified and purified.The virus titer of recombinant adenovirus was determined.AD11-RGD-4C-EGFP was used to infect HepG2,BEL-7404,and BJ cell lines,and their transduction efficiency was determined by fluorescence microscope.ResultsA 1 123 bp target gene fragments was obtained by PCR from the recombinant adenovirus; meanwhile,we prepared high titer recombinant adenovirus,with the titer being 1.3×1010pfu/ml after purification.At 10 MOI,the infection efficiency of recombinant adenovirus was much higher than control adenovirus(AD11-EGFP) after 48 h infection.ConclusionWe have successfully constructed a chimeric adenovirus harboring RGD-4C,which paves a way for enhancing the infection efficiency of adenovirus in bio-therapy.

Abstract:ObjectiveTo summarize our initial experience on single-incision laparoscopic ureterolithotomy in 6 patients, and to evaluate the efficacy and safety of this procedure. MethodsFrom Jun. 2009 to Jul. 2009, single-incision laparoscopic ureterolithotomies were performed in 6 patients who had a ureteral calculus. The clinical data and operative results were summarized retrospectively. ResultsAll 6 operations were performed successfully, with no conversion to an open procedure; neither an extra trocar for assistance was needed. No intraoperative complications were noticed. The average operative time was (93.5±30.4) min, the average estimated blood loss was (28.5±16.2) ml, and the average postoperative hospital stay was (5.4±1.1) d. ConclusionSingle-incision ureterolithotomy appears to be safe and effective, and has less trauma and better cosmetic outcomes; but it seems to be more difficult for the surgeon to operate.

Abstract:ObjectiveTo investigate the surgical diagnosis and treatment of chronic stenosis of mechanical prosthetic valve complicated with acute dysfunction, so as to deepen our knowledge on chronic stenosis of mechanical prosthetic valve. MethodsThe clinical data of 5 patients with chronic stenosis of mechanical prosthetic valve complicated with acute dysfunction were retrospectively analyzed, and the relevant literatures were reviewed. ResultsRe-operation (mechanical prosthetic valve replacement) was performed once the diagnoses were confirmed. The patients recovered well; the cardiac function was obviously improved; and there were no early complications. ConclusionChronic stenosis of mechanical prosthetic valve should be strongly suspected when they have symptoms indicating valvular stenosis. Complication of acute mechanical prosthetic valve dysfunction is not difficult to diagnose, and prompt operation is important to save the life of patients.

Abstract:ObjectiveTo isolate and identify the chemical constituents of the EtOAc fraction of Tithonia diversifolia.MethodsThe dried aerial parts of Tithonia diversifolia were extracted with 80% EtOH,and the chemical constituents of the ethyl acetate portion from the extract were isolated by column chromatography with silica gel, Sephadex LH-20 and MCI.The isolated compounds were identified by NMR and MS analysis.ResultsTen compounds were isolated from EtOAc extract, and identified as:vanillin(1), ergosterol(2), ergosterol peroxide(3), 3-methoxy-4-hydroxy-trans-cinnamaldehyde(4), p-hydroxybenzaldehyde(5), 7-methylesculetin(6), 5-hydyoxymethyl furfural(7), nepetin(8), docosanoic acid 2, 3-dihydroxypropyl ester(9) and 13-hydroxyeupatorochromene(10).ConclusionVanillin, ergosterol, ergosterol peroxide, 3-methoxy-4-hydroxy-trans-cinnamaldehyde, p-hydroxybenzaldehyde, 7-methylesculetin, 5-hydyoxymethyl furfural, docosanoic acid 2, 3-dihydroxypropyl ester, and 13-hydroxyeupatorochromene have been isolated from Tithonia diversifolia for the first time.

Abstract:ObjectiveTo establish a reversed-phase ion-pair chromatography (RP-IPC) method for determination of sinomenine content in Sinomenium acutum. MethodsThe IPC method was established using a column of Eclipse Plus C18 (4.6 mm×250 mm, 5 μm) with a mobile phase of acetonitrile and buffer phosphate (0.03 mol/L potassium dihydrogen phosphate and 0.03 mol/L sodium heptane sulfonic acid,adjusted to pH 4.0 by phosphoric acid)1882.The flow rate was 1.0 ml/min; the detection wavelength was set at 262 nm, and the temperature was at 40℃. ResultsThe linearity was obtained over a concentration range of 0.025-0.5 mg/ml(r=1.000). The low limit of detection was 2.5 μg/ml. The intra-day precisions of low, medium, and high concentrations of sinomenine were 1.54%, 0.70%, and 0.45%, and the inter-day precisions were 1.54%,0.70%, and 0.45%, respectively.The average recovery was 99.9%(RSD=2.9%, n=6), which met the requirement for analysis. Conclusion The present method is simple, fast, and accurate; it is suitable for determination of sinomenine content in Sinomenium acutum.

Abstract:Aging as a global problem is intensifying with the development of economy; reducing the chronic diseases in the elderly and improving the life quality of the whole population are a study focus of medical researchers of today.The changes of cells,tissues,and organs in adults are not reversible to different extents.Fetal origin hypothesis believes malnutrition early in life is closely associated with the metabolic syndrome in adults,and if measures is taken at early stage of life,it may reduce or avoid a series of disease in adults.This paper reviews the association of malnutrition at early stage of life with metabolic syndrome (obesity,hyperglycemia,hypertension,and hyperlipemia) in adults.

Abstract:ERCP has become an important method for diagnosis and treatment of digestive diseases, but the high incidences of post-ERCP complications (PEP), especially for post-ERCP pancreatitis, have limited its application. Prevention of PEP has become a focus of ERCP-related studies.Current drug prevention of PEP is not satisfactory, and there has not been a single drug which can effectively prevent PEP. In contrast, non-pharmacologic preventive techniques such as pancreatic stent placing and guide wire cannulation have been proven to have prominent effects in preventing PEP, which brings new hopes for PEP prevention. This paper reviews the studies on non-pharmacologic prevention of PEP.

Abstract:Endogenous hydrogen sulfide is the third gaseous signal molecule after nitric oxide and carbon monoxide; it is synthesized through cystathionine-β-synthase and cystathionine-γ-cleavage pathway.Hydrogen sulfide can directly react with the KATP channel, reduce extracellular Ca2+ influx, and work together with testosterone, nitric oxide, carbon monoxide, relaxing corpus cavernosum smooth muscle and improving diastolic function of blood vessels.It can also inhibit vascular smooth muscle cell proliferation, promote apoptosis, and improve vascular remodeling.Therefore, in-depth study of H2S in the regulating penile erection can provide a new way for the treatment of erectile dysfunction.

Abstract:目的探讨microRNA-196a2 T>C基因多态性与肝硬化发生的相关性。方法采用聚合酶链反应和连接酶检测反应法(PCR-LDR)检测110例肝硬化患者及391例健康对照者 microRNA-196a2 T>C基因多态性,运用SPSS 10.0软件对结果进行统计学分析,并分别对男性及女性个体microRNA-196a2 T>C基因多态性进行分析。结果肝硬化组与对照组都是以CT基因型为主,CC基因型及等位基因C在肝硬化组的分布频率(0.191,0.423)低于健康对照组(0.235,0.487),但无统计学差异(P>0.05);该位点基因型及等位基因在男性或女性个体中的分布趋势与整体趋势相似, 相关差异也无统计学意义(P>0.05)。结论在中国汉族人群中,microRNA-196a2 T>C基因多态性与肝硬化的发生并没有明显相关性。

Abstract:目的研究中药方剂Bingdeling体外对金黄色葡萄球菌等细菌的抑制作用。方法采用菌落形成计数方法,观察Bingdeling对临床常见4种细菌 (金黄色葡萄球菌、大肠杆菌、沙门菌、铜绿假单胞菌)体外抑菌作用;采用液体培养基连续稀释法测定Bingdeling体外对金黄色葡萄球菌生长的最小抑菌浓度(MIC);采用纸片扩散法进行中药方剂Bingdeling联合青霉素以及β-内酰胺类抗生素(阿莫西林)、氨基糖苷类抗生素(庆大霉素)、大环内酯类抗生素(罗红霉素)等抗菌药体外对金黄色葡萄球菌的药敏试验。结果体外抑菌实验结果表明,中药方剂Bingdeling体外对金黄色葡萄球菌的生长具有非常明显的抑制作用;Bingdeling体外对金黄色葡萄球菌生长的MIC是原液的0.016倍;体外Bingdeling联合青霉素、阿莫西林、庆大霉素、罗红霉素应用,对金黄色葡萄球菌有协同增强抗菌作用。结论Bingdeling体外对金黄色葡萄球菌有明显的抑菌作用,且与阿莫西林等抗菌药有协同抑菌作用,具有潜在的临床应用价值。

Abstract:目的探讨HMGA2在原发性肝癌中的表达及其与原发性肝癌患者临床资料间的关系。方法通过RT-PCR检测HMGA2 mRNA在肝癌组织及正常肝组织中的表达,蛋白质印迹分析、免疫组化分别检测HMGA2蛋白在肝癌组织及正常肝组织中的表达水平,分析HMGA2蛋白的表达与肝癌患者临床资料之间的相关性。结果 HMGA2 mRNA在肝癌组织中的阳性表达率为43.69%,高于在正常肝组织中23.30%的阳性表达率,其在肝癌组织中的相对表达值为0.71±0.03,高于其在正常肝组织中0.28±0.04的相对表达值 (P<0.05)。 HMGA2蛋白在肝癌组织中的相对表达值为0.56±0.04,高于其在正常肝组织中0.19±0.03的相对表达值 (P<0.05)。免疫组化结果显示HMGA2蛋白在正常肝组织中阳性表达率为20.39%,明显低于其在肝癌组织中37.86%的阳性表达率 (P<0.05),HMGA2蛋白的表达与肝癌患者性别和肿瘤大小无关(P>0.05),但与淋巴结转移数量、肝内转移以及临床分期相关 (P<0.05或P<0.01)。结论HMGA2在肝癌组织中高表达,且其高表达与肝癌的淋巴结转移数量、肝内转移及临床分期紧密相关,可作为肝癌转移和恶化的肿瘤标志物。

Abstract:目的观察碘缺乏大鼠肾脏的损伤情况及核转录因子激活蛋白-1(activator protein-1,AP-1)亚单位c-Jun在肾组织中的表达差异,探讨低碘地区肾脏损伤的可能发病机制。方法给予Wistar大鼠缺碘饲料,通过饮用含不同浓度碘的水分为重度缺碘组、轻度缺碘组和正常对照组,3组每日总碘摄入量分别约为1.24、5、10 μg。测定甲状腺功能状态,利用H-E染色观察肾脏病理变化,利用RT-PCR、免疫组化方法分析肾脏组织中AP-1亚单位c-Jun在基因和蛋白水平表达的差异。结果重度缺碘组与轻度缺碘组、对照组比较TT3、TT4、FT3、FT4均明显下降(P<0.01),轻度缺碘组与对照组相比TT4、 FT3明显下降(P<0.01),TT3、FT4稍有降低但差异无统计学意义。缺碘大鼠肾小球系膜区扩大,肾小球面积减小,随缺碘程度增加而加重。对照组大鼠肾组织可少量表达c-Jun,重度缺碘组与轻度缺碘组大鼠肾组织中c-Jun在基因及蛋白水平表达均增强,且重度缺碘组与轻度缺碘组比较差异有统计学意义(P<0.01)。结论碘缺乏大鼠甲状腺激素分泌减少,肾脏发生损伤,肾组织c-Jun表达上调。

Abstract:目的比较乳酸林格液(LR)和6%羟乙基淀粉200/0.5(HES 200/0.5)对脓毒症犬进行液体复苏时对氧代谢和毛细血管通透性的影响,为液体选择提供依据。方法30只犬随机分为3组(n=10):对照组行假手术后用乳酸林格液作为输注液体,LR组造脓毒症模型后用乳酸林格液作为复苏液体,HS组造脓毒症模型后用羟乙基淀粉200/0.5作为复苏液体。在脓毒症前、脓毒症后1、4、8 h测白蛋白逃离率(AER)、氧耗(VO2)、氧供(DO2)、混合静脉氧饱和度(SvO2)、心排血量(CO)、心脏指数(CI)、血浆容量(PV)、动脉血压(MAP)、中心静脉压(CVP)和肺毛细血管楔压(PCWP)等。结果 LR、HS组MAP、CO、 DO2 、SvO2在脓毒症发生后4、8 h较同组基础值及对照组均有明显下降,但HS组较LR组高(P<0.05);LR组的VO2在脓毒症发生后8 h较基础值明显下降(P<0.05),而对照组和HS组VO2无明显变化;在脓毒症发生后4、8 h,对照组血浆容量(PV)明显升高,LR组明显降低(P<0.05),而HS组无明显变化;在脓毒症发生后4、8 h,LR、HS组AER较基础值及对照组明显升高(P<0.05),而LR和HS组间无明显差异。结论脓毒症后毛细血管通透性增加,乳酸林格液、 6%HES对犬脓毒症后毛细血管通透性的影响没有明显差异,但6%HES更有利于血浆容量的维持和氧合的改善。

Abstract:目的观察以腋窝淋巴结肿大为首发症状的隐匿性乳腺癌(OBC)保全乳房治疗的疗效、并发症及预后。方法回顾性分析2000年1月至2003年12月7例以腋窝淋巴结肿大为首发症状OBC保乳治疗患者的临床资料,观察其短期疗效、并发症及预后。结果仅行腋窝淋巴结清扫的7例OBC 5年总生存率为100%,同侧乳腺无癌肿,无远处转移。结论对以腋窝淋巴结肿大为首发症状的OBC华人患者,仅行腋窝淋巴结清扫的保全乳房治疗可能是有效的治疗方法。

Abstract:牛磺酸具有多种生物学功能,近来的研究热点集中于其对肝脏损伤的保护作用,尤其是对肝纤维化的防治\[1-2\]。而肝星状细胞的活化是导致肝纤维化的重要原因。本实验应用基因芯片技术筛选牛磺酸作用于肝星状细胞后的差异表达基因,并对这些基因进行功能分类和代谢通路分析,进一步阐明牛磺酸抗肝纤维化的作用机制。

Abstract:孕妇血浆中胎儿游离DNA、RNA的发现使无创产前诊断成为可能\[1\]。利用母体血浆中的胎儿游离DNA可获得胎儿的遗传信息,包括胎儿性别、血型 Rh(D)状态等\[2\];母体血浆胎儿RNA的发现也为应用胎儿游离核酸无创进行产前诊断提供了条件\[3\]。MicroRNAs (miRNAs)是一种大小约21~23个碱基的单链小分子RNA,可通过与靶基因非编码区的结合来抑制翻译,从而调控基因表达,参与一系列生命进程,包括早期发育、细胞增殖、细胞凋亡和细胞分化等\[4\]。 母体血浆胎源性miRNAs的发现为无创性产前诊断开辟了一个新视角\[5\]。miRNA通过与一些小分子微颗粒结合免于被RNases降解,而稳定存在于孕妇血浆中\[6\],有利于无创性产前诊断;但由于其含量甚微且对保存温度要求较高,后续检测工作难度较大。因此,必须制定合理的孕妇血浆处理规范,以保证从血浆中稳定获得高浓度miRNA进行无创产前诊断。本研究观察mirVana miRNA Isolation Kit(Applied Biosystems)和TRIzol抽提法提取血浆中胎盘特异性has-miR-141的效果及稳定性,比较两者对血浆中miRNA的富集效率。

Abstract:原发性支气管肺癌是目前国内和全球发病率及病死率较高的恶性肿瘤之一,70%~80%的肺癌患者就诊时已属于中晚期而丧失手术机会\[1-3\],这些患者中约30%后期会逐渐出现肿瘤性气道阻塞。肿瘤性气道阻塞是支气管肺癌患者晚期的致死性危急重症,如不及时解除,患者很快就会因呼吸衰竭而死亡。 Vergnon等\[4\]研究认为,气道阻塞达50%以上时支气管镜介入治疗应作为首选,在镜下去除肿瘤并放置支架,然后联合放疗和化疗,可获得较好的效果。我们近年采用纤维支气管镜下高频电刀烧灼术后放置网状支架联合气管腔内放疗,对16例支气管肺癌所致的肿瘤性气道阻塞进行治疗,并观察近期疗效,总结相关经验。
