肝内胆管癌中E-cadherin/P120的表达异常

摘要:
目的:探讨肝内胆管癌中P120的表达特征及其与肿瘤临床病理特征和生存预后之间的关系。方法:应用免疫组织化学EnVision方法对42例肝内胆管癌标本进行E-cadherin和P120抗体染色。结果:E-cadherin和P120的表达减低率分别为64.3%(27/42)和73.8%(31/42)。E-cadherin和P120的表达均与肿瘤的组织学分级(分别为P=0.009和P=0.003)、TNM分期(分别为P=0.035和P:0.004)、肝内转移(分别为P=0.007和P=0.041)以及患者的生存预后(分别为P=0.024和P=0.004)之间存在显著相关。另外,P120的表达缺失与E-cadherin的表达缺失之间协调一致。Cox回归模型分析证实,只有P120和肿瘤大小是肝内胆管癌患者生存预后的独立预测因素。结论:肝内胆管癌的发生发展中常发生E-cadherin和P120的表达下调,且与肝内胆管癌的发生发展有关。二者均是肿瘤侵袭和患者预后的有价值预测指标,但只有P120是肝内胆管癌患者生存预后的独立预测因素。[著者文摘]

Altered expression of E-cadherin/P120 complex in human intrahepatic cholangiocarcinomas: correlation with clinicopathologic parameters and patients' survival

Abstract:
Objective:To investigate the expression of P120 in intrahepatic cholangiocarcinomas (ICCs) and its correlation with the clinicopathologic parameters and patients' survival. Methods: An immunohistochemical study was performed using Dako EnVision Kit to detect the expression of E-cadherin and P120-catenin in 42 ICC specimens. Results.. The expression of E-cadherin and P120 was reduced in 27 (64. 3%) specimens and 31 (73. 8%) specimens, respectively. The expression of E-cadherin and P120 was significantly correlated with the tumor histological grade (P= 0. 009 and P= 0. 003, respectively), pTNM stage (P=0. 035 and P= 0. 004, respectively), intrahepatic metastasis (P=0. 007 and P= 0. 041, respectively) , and patients'survival periods (P = 0. 024 and P= 0. 004,respectively). In addition, the expression oI P120 accorded with that of E-cadherin (P〈0. 001). We also found that P120 expression and the tumor size were the independent prognostic factors for ICC patients by a Cox regression model. Conclusion: Down-regulation of E-cadherin and P120 expression occurs frequently in ICC patients, which might be associated with the progression and development of ICC. Both of them may be valuable biologic markers for predicting tumor invasion, metastasis and patients' survival, but only P120 was an independent prognostic factor for patients' survival.[著者文摘]
