选择性环氧合酶-2抑制剂NS-398体外抑制结肠癌细胞增殖和侵袭

摘要:
目的:观察环氧合酶-2(COX-2)抑制剂NS-398对结肠癌细胞增殖、侵袭的影响,探讨COX-2作为结肠癌治疗靶标的可行性。方法:用RT-PCR和Western印迹法检测结肠癌细胞CW-2、COLO-320中COX-2基因及蛋白的表达。MTT法观察NS-398对COLO-320细胞增殖的抑制作用;体外细胞侵袭试验检测NS-398对细胞侵袭能力的影响;Western印迹检测NS-398对基质金属蛋白酶2(MMP-2)表达的影响。结果:结肠癌细胞株CW-2、COLO-320中COX-2均有阳性表达。NS-398能抑制结肠癌细胞株COLO-320的生长,抑制作用具有时间、浓度依赖性。细胞侵袭试验表明,NS-398体外能抑制结肠癌细胞株COLO-320的侵袭。Western 印迹结果表明NS-398可以抑制COLO-320中MMP-2的表达。结论:COX-2抑制剂NS-398体外可显著抑制结肠癌细胞的生长、侵袭,COX-2可作为结肠癌治疗的靶点。
