乳腺癌组织中N-乙酰氨基葡萄糖转移酶Ⅴ的活性和临床意义

摘要:
目的:检测乳腺癌组织中N乙酰氨基葡萄糖转移酶Ⅴ(GnTⅤ)的活性并分析其临床意义。方法: 应用组织芯片技术构建12×10乳腺癌组织阵列块,通过凝集素印迹法检测以白细胞植物凝集素(PHAL)标记的GnTⅤ在57例乳腺癌和 12 例乳腺良性病变中的活性,并探讨其与临床病理学参数间的关系。结果: 乳腺癌组织中PHAL阳性89.47%(51/57),胞质阳性,乳腺良性病变组织中PHAL阳性8.33%(1/12),两者存在显著性差异(P<0.01)。乳腺癌中GnTⅤ活性在不同病理类型、不同淋巴结转移状况、不同病理分期患者中的表达无明显差异。结论: 乳腺病灶GnTⅤ高活性提示病变恶性可能大,可以GnTⅤ为靶点,治疗出现复发及转移三阴的乳腺癌患者。

Abstract:
Objective:To explore the activity of Nacetylglucosaminyltransferase Ⅴ(GnTⅤ) in breast cancer tissues and to discuss its clinical significance.Methods:The activities of GnTⅤwere examined in a tissue microarray cohort of 57 breast carcinomas and 12 benign breast lesions.Lectin blotting using Phaseolus vulgaris leukocytic phytohemagglutinin (PHAL) labeling was used to assay GnTⅤactivity.The relationship between clinicopathological parameters and GnTⅤactivity was investigated.Results:High activity of GnTⅤwas observed in 89.47% (51/57) breast cancer tissues,which was significantly higher than that in the benign breast lesions (8.33%\[1/12\],P<0.01).No difference in GnTⅤactivities was observed in breast cancer patients with different pathological types,different lymphatic statuses,and different stages.Conclusion:Higher GnTⅤactivity in breast cancer tissues indicates a higher risk of malignancy.Targeting GnTⅤmight be a future way for treatment of triplenegative breast cancer patients with recurrence and metastasis.
