FoxM1和COX-2在鼻咽癌组织中的表达及临床意义

摘要:
目的 研究叉头框M1(forkhead box M1,FoxM1)转录因子和环氧合酶2(cyclooxygenase-2,COX-2)在鼻咽癌组织中的表达并探讨其临床意义。方法 运用免疫组织化学方法检测80例鼻咽癌组织和40例鼻咽部正常组织中FoxM1和COX-2的表达,分析其与鼻咽癌临床病理参数的关系,以及FoxM1和COX-2表达之间的关系。结果 FoxM1和COX-2在鼻咽癌组织的阳性表达率均高于鼻咽部正常组织\[FoxM1: 68.8% (55/80) vs 2.5% (1/40); COX-2: 62.5% (50/80) vs 5.0% (2/40); P<0.05\]。FoxM1阳性表达与淋巴结转移(rs=0.252,P<0.05)和临床分期(rs=0.230,P<0.05)相关;COX-2阳性表达与淋巴结转移(rs=0.287,P<0.05)和临床分期(rs=0.239,P<0.05)相关。FoxM1 与COX-2表达呈正相关(rs=0.631,P<0.05),提示二者可能有直接或间接作用关系。结论 FoxM1与COX-2在鼻咽癌组织中表达的上调可能与鼻咽癌的发生及浸润转移有关。

Abstract:
Objective To investigate the expression of forkhead box M1(FoxM1) transcription factor and cyclooxygenase-2 (COX-2) in nasopharyngeal carcinoma (NPC) tissues, and to discuss the related clinicopathological implications. Methods Immunohistochemistry method was used to examine the expression of FoxM1 and COX-2 in 80 NPC tissues and 40 normal nasopharyngeal mucosa tissues. The relationship of FoxM1 and COX-2 expression with the clinicopathological parameters of NPC was analyzed; we also analyzed the correlation between the FoxM1 and COX-2 expression. Results FoxM1 and COX-2 expressions were significantly higher in NPC tissues compared to normal nasopharyngeal mucosa tissues (FoxM1: 68.8% \[55/80\] vs 2.5% \[1/40\]; COX-2: 62.5% \[50/80\] vs 5.0% \[2/40\]; P<0.05). The expression of FoxM1 was significantly associated with lymph node metastasis (rs=0.252, P<0.05) and clinical stage of NPC (rs=0.230, P<0.05), and expression of COX-2 was also significantly associated with lymph node metastasis (rs=0.287, P<0.05) and clinical stage of NPC (rs=0.239, P<0.05). Furthermore, a positive correlation was observed between FoxM1 and COX-2 expression (rs =0.631, P<0.05), indicating a direct or indirect interaction between them. Conclusion Increased expression of FoxM1 and COX-2 in NPC tissues might be associated with the development and progression of NPC.
