目的:考察Graves病患者血浆睾酮(testosterone,T)浓度以及其外周血白细胞和甲状腺组织中雄激素受体(androgen receptor,AR)水平,以探讨雄激素及其受体与Graves病的关系.方法:以8例女性Graves病患者、8例甲状腺腺瘤患者及8例配对的健康对照者为研究对象,采用酶联免疫分析法测定血浆T浓度,放射配体结合法检测患者外周血白细胞3H雄激素(T)特异结合位点,RT-PCR检测外周血白细胞AR的mRNA表达,免疫组织化学法检测甲状腺组织AR水平.结果:Graves病组和对照组血浆T浓度分别为(660±80) ng/L及(620±100) ng/L,组间无明显差异(P>0.05);两组外周血白细胞 3H-T特异结合位点分别为(381±105)位点/细胞、(572 ±141)位点/细胞,组间差异显著(P<0.05);Graves病患者外周血白细胞AR的mRNA表达量显著高于对照组(P<0.05);免疫组化检测显示,Graves病患者甲状腺组织AR水平显著高于正常甲状腺组织(P<0.01).结论:Graves病患者血浆T浓度较正常对照组无明显变化,但外周血白细胞AR的mRNA表达和甲状腺组织中AR水平均增高,考虑是机体保护性反应的调节.此外,也可能与病理情况下性激素结合能力下降、AR不能良好发挥生理效应相关.外周血白细胞中蛋白结合活性检测结果降低,推测是甲亢患者AR分解加速所致.[著者文摘] |

Expression of androgen receptor gene in thyroid tissues of patients with Graves disease |

Objective: To observe the serum testosterone (T) level and androgen receptor (AR) mRNA expression in peripheral leukocytes and thyroid tissues of patients with Graves disease, so as to investigate the relationship between the testosterone and androgen receptor with hyperthyroidism. Methods: Blood samples were collected from 8 female patients with Graves disease,8 female patients with thyroid adenoma and 8 healthy control subjects; the patients were paired in gender and age. The plasma concentrations of testosterone were determined by enzyme-linked immunosorbent assay (ELISA) ; the [^3 H]-T specific binding sites in the peripheral leukocytes were determined by radioligand binding assay; the expression of AR mRNA in peripheral leukocytes were determined by RT-PCR; and the level of AR in thyroid tissues was determined by immunohistochemistry method. Results: The plasma concentrations of T were (660±80) ng/L and(620± 100) ng/L in Graves disease group and control group, respectively (P〉 0. 05) ; the [^3 H]-T specific binding sites were ( 381 ± 105) sites/cell and (572 ± 141 ) sites/cell in peripheral leukocytes, respectively ( P〈 0.05 ). The mRNA expression of AR in Graves disease group was significantly higher than that in the control group(P〈0.05). The result of immunohistochemistry showed that the level of AR in thyroid tissues was higher in Graves group than that in the control group. Conclusion: The plasma level of T in patients with Graves disease is similar to that of normal controls, but the mRNA expression of AR in peripheral leukocytes and thyroid tissue is higher in Graves disease group than in the control group, which is supposed to be a protective reaction. Moreover, the binding capability of AR is lower in Graves disease group than that in the control group, which is probably caused by the accelerated decomposition of AR.[著者文摘] |
