乳腺癌组织中N-乙酰氨基葡萄糖转移酶Ⅴ的活性和临床意义 |

摘要: |
目的:检测乳腺癌组织中N乙酰氨基葡萄糖转移酶Ⅴ(GnTⅤ)的活性并分析其临床意义。方法: 应用组织芯片技术构建12×10乳腺癌组织阵列块,通过凝集素印迹法检测以白细胞植物凝集素(PHAL)标记的GnTⅤ在57例乳腺癌和 12 例乳腺良性病变中的活性,并探讨其与临床病理学参数间的关系。结果: 乳腺癌组织中PHAL阳性89.47%(51/57),胞质阳性,乳腺良性病变组织中PHAL阳性8.33%(1/12),两者存在显著性差异(P<0.01)。乳腺癌中GnTⅤ活性在不同病理类型、不同淋巴结转移状况、不同病理分期患者中的表达无明显差异。结论: 乳腺病灶GnTⅤ高活性提示病变恶性可能大,可以GnTⅤ为靶点,治疗出现复发及转移三阴的乳腺癌患者。 |

Objective:To explore the activity of Nacetylglucosaminyltransferase Ⅴ(GnTⅤ) in breast cancer tissues and to discuss its clinical significance.Methods:The activities of GnTⅤwere examined in a tissue microarray cohort of 57 breast carcinomas and 12 benign breast lesions.Lectin blotting using Phaseolus vulgaris leukocytic phytohemagglutinin (PHAL) labeling was used to assay GnTⅤactivity.The relationship between clinicopathological parameters and GnTⅤactivity was investigated.Results:High activity of GnTⅤwas observed in 89.47% (51/57) breast cancer tissues,which was significantly higher than that in the benign breast lesions (8.33%\[1/12\],P<0.01).No difference in GnTⅤactivities was observed in breast cancer patients with different pathological types,different lymphatic statuses,and different stages.Conclusion:Higher GnTⅤactivity in breast cancer tissues indicates a higher risk of malignancy.Targeting GnTⅤmight be a future way for treatment of triplenegative breast cancer patients with recurrence and metastasis. |
