\[摘要\]目的利用体外培养的大鼠乳鼠心肌细胞,探讨葛根素(puerarin,Pue)对异丙肾上腺素(isoproterenol,ISO)所致心肌细胞肥大和心肌细胞凋亡的影响。方法新生大鼠心肌细胞培养,除对照组外,分别给予ISO(10-5mol/L)、Pue(10-6mol/L)、ISO(10-5mol/L)+Pue(10-6mol/L)、ISO(10-5mol/L)+普萘洛尔(propranolol,Pro, 2×10-6 mol/L),检测培养心肌细胞蛋白质含量、细胞体积、蛋白质的合成及细胞凋亡率。结果和对照组相比,ISO可使心肌细胞总蛋白含量、体积和凋亡率明显增加(P<0.01);Pue可以抑制由于ISO诱导所出现的上述情况,结果与Pro相近。结论Pue能对抗ISO引起的心肌肥大及细胞凋亡,此效应可能是葛根素抑制心肌肥厚发生的保护机制之一。 |

\[Abstract\]ObjectiveTo study the effect of puerarin (Pue) on hypertrophy and apoptosis of neonatal rat cardiomyocytes induced by isoproterenol(ISO). MethodsCardiomyocytes of neonatal rats were cultured in vitro and were treated with ISO(10-5mol/L), Pue(10-6mol/L), ISO(10-5mol/L)+ Pue(10-6mol/L), or ISO(10-5mol/L)+propranolol (Pro, 2×10-6 mol/L); untreated cells were taken as control. The total protein content, cardiomyocyte volume, protein synthesis and apoptosis rate of the cardiomyocytes were investigated. ResultsCompared with the control group, ISO significantly increased the total protein content, cardiomyocyte volume and apoptosis rate of cardiomyocytes (P<0.01).Similar to Pro, Pue inhibited the above changes induced by ISO. ConclusionPue can inhibit the hypertrophy and apoptosis of cardiomyocytes induced by ISO, which might be one of the reasons for the protective effect of Pue against myocardial hypertrophy. |
