DJ-1抗氧化应激功能研究进展

摘要:
氧化应激涉及多种疾病的发生和发展以及人体的衰老过程。DJ-1/PARK7基因编码DJ-1蛋白,研究发现其具有多种生物学功能,包括细胞的恶性转化、雄性精子成熟与受精、促进细胞增殖及抵抗氧化应激等。近年来DJ-1抗氧化应激功能日益受到重视。研究层次包括体外实验、体内实验和临床研究。其机制主要包括自身构型改变、分子伴侣作用、保护线粒体功能、调控抗凋亡基因和抗氧化相关基因。对DJ-1进一步研究可能为氧化应激相关性疾病的预防和治疗提供新策略。

Abstract:
Oxidative stress is implicated in the pathogenesis and progression of various diseases as well as in the natural aging process. DJ-1/PARK7 gene encodes DJ-1 protein, which participates in malignant transformation, sperm maturation and fertilization, cell proliferation promotion, and anti-oxidative stress. The anti-oxidative stress effect of DJ-1 has been drawing increasing attention in recent years. There have been related in vitro, in vivo and clinical studies. The main mechanisms of its anti-oxidation effects included modifying configuration, playing the role of molecular chaperones, protecting the mitochondrial function, and regulating anti-apoptosis genes and antioxidant-related genes. Further research on DJ-1 may provide a new strategy for the prevention and treatment of oxidative stress-related diseases.
