多西紫杉醇联合环巴胺抑制脑胶质瘤生长的实验研究

摘要:
目的 研究多烯紫杉醇(DOC)联合Hh信号通路拮抗剂环靶明对脑胶质瘤的治疗效果。方法 用MTT法考察多烯紫杉醇联合Hh信号通路拮抗剂环靶明对脑胶质瘤细胞的增殖抑制情况。构建U87脑胶质瘤细胞肿瘤球模型,考查联合给药对肿瘤球体积的增长抑制能力。构建脑胶质瘤裸鼠异位肿瘤模型,随机分组,考察联合给药对脑胶质瘤的肿瘤生长的抑制效果,并记录荷瘤裸鼠的生存期。结果 相比于单独用药组,联合用药对肿瘤细胞的增殖抑制作用和对肿瘤球的生长抑制作用显著强于单独用药。通过异位肿瘤模型证实,联合用药能够显著抑制肿瘤体积生长,延长荷瘤裸鼠的中位生存期。结论 多烯紫杉醇与环靶明均能抑制脑胶质瘤的生长,二者联合使用能够发挥协同作用,是一种潜在的高效的脑胶质瘤治疗手段。

Abstract:
Objective:To estimate the effect of docetaxel combined with cyclopamine on the treatment of glioma. Methods The anti-proliferation efficiency of DOC combined with cyclopamine was evaluated by MTT assay. Tumor spheroids were used to evaluate anti-tumor ability of DOC DOC combined with cyclopamine. U87 cells were xenografted in athymic nude mice to establish the animal model, which were used to evaluate the effect of anti-glioma. Results The anti-proliferative activity against U87s and tumor spheroids confirmed strong inhibitory effect of docetaxel combined with cyclopamine. The combination of DOC and cyclopamine can prolong the median survival time of U87 bearing nude mice than other groups significantly. Conclusion the DOC and cyclopamine can inhibit the growth of gioma, and the combination of DOC and cyclopamine may be a potentially effective treatment for glioma.
