目的 检测钠耦合中性氨基酸转运子1(SNAT1)在食管鳞癌组织中的表达,分析其在评估食管鳞癌患者预后中的价值。方法 构建含183例食管鳞癌和癌旁组织的组织芯片,用免疫组化技术和蛋白质印迹法检测SNAT1蛋白在食管鳞癌组织中的表达,并分析其与临床病理参数和总生存时间的相关性。结果 SNAT1在正常鳞状上皮组织中不表达,在食管鳞癌组织中高表达,过表达率为45.4%(83/183);SNAT1的过表达与肿瘤大小(P=0.023)、淋巴结转移(P=0.007)以及疾病进展(P=0.003)有关;生存分析显示SNAT1过表达患者的预后要劣于SNAT1阴性表达患者(P<0.001)。此外,SNAT1过表达是评估食管鳞癌患者预后的独立因素(P=0.004)。结论 SNAT1在食管鳞癌中异常活化,是判断食管鳞癌患者预后的重要因素之一。 |

Abstract: |
Objective To investigate the expression of sodium-coupled neutral amino acid transporter 1 (SNAT1) in esophageal squamous cell carcinoma (ESCC) tissues, and to evaluate its prognostic value in patients with ESCC. Methods The tissue microarray including 183 ESCC tissues and their corresponding adjacent tissues was subjected to immunohistochemistry detection and Western blotting analysis of SNAT1 expression, and their correlation with the clinicopathological parameters and overall survival time was analyzed. Results SNAT1 was highly expressed in ESCC tissues, with the positive rate being 45.4% (83/183); we also noticed that it was not expressed in normal squamous epithelium tissues. Our analysis results indicated that overexpression of SNAT1 was significantly correlated with tumor size (P=0.023), lymph node metastasis (P=0.007) and tumor stage (P=0.003). The Kaplan-Meier analysis showed that patients with overexpression of SNAT1 had significantly worse outcome than those with negative SNAT1 expression (P<0.001). Cox regression analysis revealed that overexpression of SNAT1 was an independent factor for prognostic of patients with ESCC (P=0.004). Conclusion The expression of SNAT1 is abnormally increased in ESCC tissue, indicating that SNAT1 expression may serve as an important biomarker for prognosis evaluation in patients with ESCC. |
